摘  要:目的建立防风通圣丸五波长(246、254、265、280、300 nm)高效液相色谱指纹图谱(HPLC-FP)、紫外光谱指纹图谱(UV-FP)、红外光谱指纹图谱(IR-FP)和热值谱(HVF),形成多元多维方法交叉定量评价FFTSP质量。方法用系统指纹定量法(SQFM)评价五波长指纹图谱并用指纹熵权整合评价FFTSP质量。用190-400 nm区间UV-FP表达π→π*,n→π*和n→σ*的化学组分,用4 000-400 cm-1 IR-FP表达单键、双键和三键等的振动(或转动)对中红外线的吸收,均以光谱点为评价单元。用燃烧焓衡量中药有机物达到高价氧化态的热释放度,从而揭示有机组分总量。结果将五波长HPLC指纹图谱、光谱指纹谱和热值谱交叉定量评价出S1质量劣(8级),其余样品质量均在中等以上。4种检测信息相互补充、相应对照,描绘出FFTSP多元化学成分的分布状况,最终鉴定S7可作为FFTSP的标准制剂和标准指纹对照物。结论此4种多元多维指纹图谱的交叉整合能监测中药各类化学成分的分布情况,为中药质量控制提供了一种简单,价廉的探索性方法。多元多维指纹图谱技术在原理上优势互补,其合理交叉整合定性定量信息具备综合鉴定中成药质量全貌的技术基础,这为寻找中成药大复方体系的标准制剂提供了一种探索模式,具有现实性、有效性、可靠性和可操作性。Objective To establish the 5-wavelength(246, 254, 265, 280 and 300 nm) HPLC fingerprint, ultraviolet fingerprint(UV-FP), infrared fingerprint(IR-FP) and the heat value fingerprints(HVF) of Fangfeng Tongsheng pills(FFTSP), in which the multi-element and multi-dimentional fingerprints were performed to evaluate FFTSP quality consistency. Methods The systematic qualified fingerprint method(SQFM) was used to assess the 5-wavelength fingerprint and their integrated fingerprint by entropy weight method to present FFTSP authentical quality. Meanwhile UV-FPs from 190 nm to 400 nm were used to reflect the information of π→π*, n→π* and n→σ* within the chemical constitution, and the IR-FPs were used to determine the intermediate infrared absorption in 4 000- 400 cm- 1 resulted from the vibration(or rotation) of the single bond, double bond and triple bond(the single bond was especially important), where each spectrum point was used as the evaluation unit by SQFM. The heat of combustion represented all the TCM chemical composition to become the higher oxidation state to overall display the total content of all organic chemical constituents. Results HPLC-FP, UV-FP, IR-FP and HVF were crosswise organically integrated to evaluate the quality of S1 as the inferior(grade 8), and the others were beyond the moderate(grade 5). The 4 kinds of determination information were supplementary and contrast to describe the distribution of all chemicals. Finally S7(20100308) compound prescription served as the standard prescription and the reference standard material for the fingerprint. Conclusion These 4 kinds of multi-element and multi-dimentional fingerprints crosswise integrated can potentially and comprehensively monitor and assess the distribution of all chemicals, which is simple and of low cost.
