摘 要:[目的]基于DNA条形码COI数据,发现地鳖(土鳖虫主要基原)内部可能的隐存多样性。[方法]对8批33个地鳖个体的线粒体COI片段进行PCR扩增及序列测定。分析COI序列单倍型,分析变异位点,运用邻接法、最大简约法分别构建分子系统发育树,结合种群(批次)内及临时种间的遗传距离发现隐存多样性。[结果]根据系统发育结果区分临时种EsPS1及EsPS2。分析得到COI基因的物种间阈值SST为5.33%。临时种EsPS1与EsPS2间的最小遗传距离为12.00%,显著大于物种间阈值SST。[结论]地鳖内部存在隐存物种多样性。[Objective] To investigate the cryptic species diversity among Eupolyphaga sinensis as one main resource of TCM Tubiechong, based on the DNA Barcoding COI sequences.[Methods] Mitochondrial COI fragments from 33 individuals of 8 batchs of Eupolyphaga sinensis were amplified and sequenced. The haplotypes of COI and then the variation locuses were analysed. The phylogenetic trees based on NJ and MP methods were constructed, and the cryptic species were discovered by the genetic distance analysis of intra-population and inter-provisional species samples. [Results] Provisional species EsPS1 and EsPS2 were distinguished by phylogenetic analysis. The species screening threshold SST of COI fragments was set as 5.33%, and the minimal genetic distance between EsPS1 and EsPS2 was 12.00%, which was higher than SST significantly. [Conclusion] There is cryptic taxonomic diversity among TCM Eupolyphaga sinensis.
