摘  要:目的 探讨七氟醚麻醉对老龄大鼠海马含GluR2亚基的α-氨基-3-羟基-5-甲基-4-异恶唑丙酸(AMPA)受体转运的影响.方法 健康雄性老龄Wistar大鼠68只,18 ~ 20月龄,体重600 ~ 650g,采用随机数字表法,将其分为2组(n=34):对照组(C组)和七氟醚组(S组).S组行开放性胫骨骨折手术,吸入3.6%七氟醚2h进行麻醉.于术后1、3、7d进行场景恐惧记忆实验、声音提示恐惧记忆实验和Y迷宫实验,采用Western blot法测定海马总蛋白及膜蛋白含GluR2亚基的AMPA受体表达,并计算膜蛋白与总蛋白该受体表达水平的比值(m/t比值).结果 与C组相比,S组术后1和3d场景恐惧记忆实验僵直时间百分比和自发交替百分比降低,海马膜蛋白含GluR2亚基的AMPA受体表达下调,m/t比值降低,术后1-7 d声音提示恐惧记忆实验僵直时间百分比降低(P<0.05).结论 七氟醚麻醉诱发老龄大鼠术后学习记忆功能障碍的机制与促进海马含GluR2亚基的AMPA受体从胞膜向胞浆转运有关.Objective To evaluate the effect of sevoflurane anesthesia on trafficking of GluR2-containing α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptors in aged rat hippocampi.Methods Sixty-eight healthy male Wistar rats,aged 18-20 months,weighing 600-650 g,were randomly divided into 2 groups (n =34 each) using a random number table:control group (group C) and sevoflurane group (group S).In group S,open tibial fracture was performed and anesthesia was induced with 3.6% sevoflurane for 2 h The contextual fear conditioning test,tone-cued fear conditioning text and Y maze test were performed on days 1,3 and 7 after surgery and the expression of GluR2 subunit-containing AMPA receptors in total and membrane proteins in hippocampi was detected by Western blot.The ratio of AMPA receptors in the membrane protein to those in the total protein (m/t ratio) was calculated.Results Compared with group C,the percentage of freezing time in the contextual fear conditioning text and percentage of spontaneous alternation were decreased,the expression of GluR2 subunit-containing AMPA receptors in the membrane protein was down-regulated,the m/t ratio was decreased on days 1 and 3 after surgery,and the percentage of freezing time in the tone-cued fear conditioning text was decreased on days 1-7 after surgery in S group (P 〈 0.05).Conclusion The mechanism by which sevoflurane anesthesia induces the impairment of learning and memory function in aged rats is related to enhanced trafficking of GluR2-containg AMPA receptors from cell membrane to cytoplasm in hippocampi.
