摘 要:目的:建立同时测定盐酸莫西沙星原料中手性异构体含量的方法。方法:采用高效液相色谱法,色谱柱为InertsilODS-3(4.6mm×250mm,5μm),以甲醇-手性添加剂溶液(含0.01mol·L^-1L-异亮氨酸和0.01mol·L^-1硫酸铜)(25:755为流动相;流速为1ml·min^-1;柱温为30℃;检测波长为293nm。结果:盐酸莫西沙星RR异构体在0.200~2.000μg·ml^-1。内、盐酸莫西沙星SR异构体在0.205-2.050μg·ml^-1内与峰面积呈良好的线性关系,r均为0.9999;平均回收率分别为98.9%o(RSD为1.17%)和98.5%(RSD为0.46)。结论:HPLC法方法简便、准确、重现性好,可作为盐酸莫西沙星原料手性异构体有关物质的质量控制方法之一。OBJECYIVE To establish a HPLC method for simultaneous determination of moxifloxacin and its isomers. METHOIIS The chromatographic method was carried out on a Inertsil ODS-3 (4. 6 x 250 mm,5μm) with methanol chiral ad- ditive solution (0. 01 mol.L-1 of L-isoleucine with 0. 01 mol·L-1 copper( II ) ) as the mobile phase. The flow rate was 1.0 ml · min^- 1 and the colurr/n temperature was 30 ℃. The detection wavelength was set at 293 nm. RESULTS The calibration curve of moxifloxacin was linear within the range of 0. 200-2. 000 tLg.ml 1 (r= 0. 999 9) for RR isomer and 01 205 -2. 050 μg.ml ^-1(r = 0. 999 9) for SR isomer,the average recoveries were 98. 9%(RSD= 1.17%) for RR isomer and 98. 5%(RSD= 0. 46%) for SR isomer. CONCLUSION The method is simple, accurate and reproducible, which can be used for the quality control of moxi floxacin hydrochloride and its chiral isomers.
