摘 要:目的 探讨白细胞介素 18(IL 18)对吡喹酮治疗后的血吸虫感染小鼠免疫功能的影响及其对虫卵肉芽肿的作用。方法 采用ELISA方法和图像分析技术 ,分析吡喹酮治疗后血吸虫感染小鼠血清IL 18、γ 干扰素 (IFN γ)、IL 4、转化生长因子 β1(TGF β1)的水平以及虫卵肉芽肿面积和羟脯氨酸水平 ,在感染后 6周 ,将小鼠IL 18重组腺病毒转染的肝细胞经脾移植到吡喹酮治疗后的感染小鼠 ,探讨IL 18对小鼠免疫功能的影响及对虫卵肉芽肿的作用。结果 吡喹酮治疗后小鼠血清IL 18、IFN γ水平明显下降 ,IL 4、TGF β1水平虽有所降低 ,但仍维持在较高水平 ;同时可见 ,虫卵肉芽肿面积和肝脏羟脯氨酸含量无明显降低 ,与同期感染组比较差异均无显著性。IL 18基因修饰的肝细胞经脾移植后能稳定表达IL 18,显著升高IFN γ水平 ,明显降低IL 4、TGF β1水平 ,减轻虫卵肉芽肿和肝纤维化程度。结论 ILObjective To investigate the immunoregulation and granuloma response in mice infected with S.japonicum following praziqual treatment by intrasplenic transplantation of IL 18 gene modified hepatocytes. Methods Six weeks after the infection of S.japonicum , the infected mice were treated by praziquantel, meanwhile, they were intrasplenic transplanted by murine hepatocytes which had been transfected with IL 18 gene combinant adenovirus vector, ELISA and profile analytical technique were used to observe the changes of serum IL 18, IFN γ, IL 4, TGF β1 level and the granuloma size. Results After praziquantel treatment serum IL 18, IFN γ level in the praziquantel treated group decreased markedly while IL 4, TGF β1 level only slight reduced, the granuloma size and hepatic hydroxyproline level did not change significantly comparing with that of infected control ( P > 0.05 ). The intrasplenic transplantation of IL 18 gene modified hepatocytes effectively expressed IL 18 and serum IFN γ level increased markedly while obviously reduced the expression of IL 4, TGF β1 as well as the granuloma size and hepatic hydroxyproline than that of infected group and praziquantel treatment group ( P <0.01). Conclusion IL 18 gene transplantation has effective function of anti hepatic fibrosis in S.japonicum infection.
