摘  要:目的探讨十全大补汤调整肉桂剂量对氧化损伤大鼠真皮成纤维细胞内活性氧水平和线粒体膜电位的影响。方法原代培养成纤维细胞,用H2O2造成氧化损伤成纤维细胞模型,以血清药理学方法获取含不同剂量肉桂的十全大补汤大鼠血清,用含药血清对成纤维细胞模型进行干预。运用流式细胞术结合DCFH-DA、JC-1探针方法检测成纤维细胞胞内活性氧和线粒体膜电位。结果含不同剂量肉桂的十全大补汤均具有良好的清除活性氧作用,组间差异无统计学意义(P>0.05)。2倍肉桂组阻滞线粒体膜电位下降明显,差异有统计学意义(P<0.01)。结论不同肉桂剂量十全大补汤可清除氧化损伤的大鼠真皮成纤维细胞胞内活性氧,2倍肉桂剂量可起到抑制细胞凋亡的作用。Objective To explore the effects of Shiquan Dabu Decoction with modified cinnamon dose on reactive oxygen and mitochondrial membrane potential in dermal fibroblasts in rats with oxidative damage. Methods The primary fibroblasts cultured in vitro was selected and turned into oxidative damage model of fibroblasts with H2 02. With serum-pharmacological method, the rats serum of Shiquan Dabu Decoction with different cinnamon doses was acquired, then the decoction-containing serum was applied to make an intervention on fibroblasts model. The flow cytometry technique and DCFH-DA, JC-1 probe method were applied to measure the active oxygen and mitochondrial membrane potential in fibroblasts. Results The Shiquan Dabu Decoction with different dose of cinnamon all had favorable effects on clearing active oxygen, without any differences among groups (P 〉 0. 05). The group with double dose of cinnamon obviously blocked the decreasing of mitochondrial membrane potential, which was statistically significant (P 〈 0. 01 ). Conclusion The Shiquan Dabu Decoction with different dose of cinnamon could clear the active oxygen in dermal fibroblasts in rats with oxidative damage, and the Decoction with double dose of cinnamon could inhibit cell apoptosis.
