摘  要:目的:评价慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)患者诱导痰T细胞巨噬细胞移动抑制因子(MIF)的表达及意义。方法:流式细胞术检测正常吸烟对照组、COPD稳定组及COPD急性加重组诱导痰中CD4+MIF+T细胞占总CD4+T细胞的百分比(CD4+MIF+T%),CD8+MIF+T细胞占总CD8+T细胞的百分比(CD8+MIF+T%),以及MIF的平均荧光强度(MFI),并与FEV1%、CAT评分等行相关性分析。结果:CD8+MIF+T%:急性加重组较稳定期升高,稳定期较对照组升高;CD4+MIF+T%、CD8+MIF+T MFI:急性加重期组较稳定期组及对照组均有明显升高,稳定期较对照组无统计学差异;CD4+MIF MFI三组无统计学差异。COPD急性加重组CD8+MIF+T%及CD4+MIF+T%与CAT评分呈正相关;稳定组CD8+MIF+T%与FEV1呈负相关,与CAT评分呈正相关。结论:MIF可能参与了COPD稳定期及急性加重期的发病。Objective To investigate the contribution of Macrophage migration inhibitory factor in patients with chronic obstructive pulmonary disease (COPD). Methods Subjects are divied into three groups: acute exacerbation of COPD(AECOPD), stable COPD(SCOPD) and normal smokers(NS). Flow cytometry was applied to measure the percentages of CD4+MIF+Tcells in the total CD4+cells(CD4+MIF+T%), CDS+MIF+Tceils in the total CD8+cells (CD8+MIF+T%), as well as detect the mean fluorescence intensity(MFI) of MIF on CD4+ and CD8+cells, correlation analysis between all of above with the lung function, the CAT score are also performed. Results CD8+MIF+T%: subjects with AECOPD was higher than SCOPD and NS, subjects with SCOPD was higher than NS; CD4+MIF+T% and MFI of MIF on CD8+MIF+T: subjects with AECOPD was higher than SCOPD and NS; In AECOPD, CD8+MIF and CD4+MIF are all positively related with lymphocytes percentage and CAT scores; in SCOPD, CD8+MIF are negatively related with FEV1, and positively related with lymphocytes percentage and CAT scores. Conclusion MIF may play an important role in the pathogenesis of COPD both stable period and acute period, The molecular mechanism remains to be further elucidated.
