沙格列汀对2型糖尿病患者血管内皮依赖性舒张功能的影响

摘　　要：目的观察二肽基肽酶-4(DPP-4)抑制剂沙格列汀对2型糖尿病患者血管内皮依赖性舒张功能(FMD)的影响。方法 80例新诊断的2型糖尿病患者随机分为沙格列汀组和对照组,各40例。沙格列汀组给予运动、饮食改善等糖尿病基础治疗同时加用沙格列汀5 mg/d,对照组给予运动、饮食等改善糖尿病基础治疗和安慰剂。治疗12周,观察两组患者治疗前后FMD、空腹血糖(FPG)、餐后2 h血糖(2 hPG)、糖化血红蛋白(HbA1C)的变化。结果治疗12周后与对照组比较,沙格列汀组FPG、2 hPG、HbA1C均显著降低(P<0.05),FMD显著升高(P<0.05)。治疗过程中沙格列汀组出现1例低血糖反应,无严重不良反应发生。结论 DPP-4抑制剂沙格列汀可以改善2型糖尿病患者血管内皮依赖性舒张功能。Objective To investigate the effect of DPP-4 inhibitor saxagliptin on endothelium-dependent flow mediated vasodilatation （FMD） in patients with type 2 diabetes. Methods A total of 80 newly diagnosed patients with type 2 diabetes were randomly divided into two groups （ n = 40 in each group）. The patients were treated with exercise - diet therapy and saxagliptin 5 mg/d in treatment group,and exercise - diet therapy and placebo in control group. Fasting blood-glucose （ FPG）, 2 h postprandial plasma glucose （ 2 hPG）, glycosylated hemoglobin A1C （HbA1 C） levels and FMD were evaluated before treatment and after 12-week treatment. Results Compared with control group, the levels of FPG,2 hPG, HbA1C were obviously decreased in treatment group （P 〈 0.05 ） and FMD were obviously increased （ P 〈 0. 05 ）. Conclusion Saxagliptin could improve the FMD in patients with type 2 diabetes.
