摘  要:目的建立二甲基甲酰胺(DMF)职业接触者血中N-甲基氨甲酰加合物(NMHb)的测定方法。方法收集DMF职业接触人群血液,用分子生物技术提取法制备成血红蛋白粉末。经降解、提取等处理后,用GCMS测定血中NMHb的降解产物3-甲基-5-异丙基乙内酰脲(MVH)的水平。结果当MVH浓度在5~300μmol/L范围内,与峰面积呈线性相关,其相关系数为0.9992,平均加标回收率为92.13%~97.44%,相对标准偏差为3.78%~4.97%,检出限和定量下限分别为0.1和0.33μmol/L。结论该方法的各项性能指标都符合GBZ/T 201.5—2008,能很好地应用于二甲基甲酰胺职业接触者血中N-甲基氨甲酰加合物的测定。Objective To establish the determination method of blood N - methylcarbamoyl adduct (NMHb) in occupational workers exposed to N, N - dimethylformamide (DMF). Methods The blood samples of occupational workers exposed to DMF were collected and made into hemoglobin powder using molecular biological extraction method. After the treatment of degradation and extraction, 3 - methyl - 5 - isopropylhydantoin ( MVH), as the degradation product of N - methylcarbamoyl adduct in blood was determined by gas chromatographic -mass spectrometric. Results When the concentration of MVH was in range of 5 - 300 I^mo//L, the relationship between concentration and peak areas of MVH was found to be linear( correlation coefficient was O, 9992). The average standard adding recovery ratio was 92. 13% -7.44%. The relative standard deviation was 3.78% -4. 97%. The detection limit and the lower limit of quantification were 0. 1μ mol/L and 0. 33 μmol/L respectively. Conclusion All performance indices of this method were in accordance with the requirement of GBZ/T 201.5 - 2008. The method could be well applied to determine blood N - methylcarbamoyl adduct in occupational workers exposed to DMF.
