摘  要:目的建立同时测定五味子酯甲、五味子甲素、五味子乙素的LC-UV含量测定方法。综合考察皖南山区野生华中五味子的内在品质,为该品种转为栽培品提供理论依据。方法采用LC-UV法,Agilent TC-C18(2)色谱柱(250 mm×4.6 mm I.D,5 um),以甲醇-水为流动相,梯度洗脱,流速为1 mL/min,柱温30℃,检测波长为240 nm,测定五味子酯甲、五味子甲素、五味子乙素。结果皖南山区野生华中五味子中五味子酯甲、五味子甲素、五味子乙素平均含量分别为0.24%、0.46%、1.22%。结论皖南山区野生华中五味子中五味子酯甲平均含量为0.24%,符合《中国药典》2010版的要求:五味子酯甲含量不低于0.20%,可供药用。通过三指标的综合控制,为皖南华中五味子的品质评价和栽培提供理论支持。Objective To investigate the determination of the contents of Schisantherin A, Deoxyschizandrin and γ-schizan- drin in S. sphenanthera Rehd. et Wils. by LC-UV and explore the quality of wild S. sphenanthera Rehd. etWils, in Wannan mountainous area to provide a reliable measure for quality control of GAP. Methods 1. LC-UV method was established with a Agilent TC-C18(2) column (5 μm,4.6 × 250 mm), a mobile phase of Methanol-Water solution a flow rate of 1.0 mL· min- 1 and a detection wavelength of 240 nm in order to determine the contents of Schisantherain A, Deoxyschizandrin and γ- schizandrin. The column temperature was maintained at 30 ℃. Results The average contents of Schisantherain A, De- oxyschizandrin andγ-schizandrin in the decoction of wild S. sphenanthera Rehd. et Wils. in Wannan mountainous area were 0.24% ,0.46%, 1.22% respectively. Conclusion The average content of Schisantherin A in wild S. sphenanthera Rehd.et Wils in Wannan mountainous area was 0.24% which is in accordance with the requirements Chinese Pharmacopoeia (2010 Edition). The content of Schisantherin A is no less than 0.20 %, which can be used for medicine. By the comprehen- sive control of three indicators, theoretical support for the quality evaluation and cultivation of S. sphenanthera Rehd. et Wils. in southern Anhui can be provided.
