紫绒与萱草杂交F_1优株快繁技术的研究

摘　　要：根据观赏性状互补原则,以引进大花萱草紫绒(Hemrocalis hybrida cv."Zirong")为父本,本地萱草属植物萱草(H.fulva)为母本进行杂交,筛选出适合哈尔滨露地应用的优秀萱草园艺品种4个,并建立了4个F1代优株的快速繁殖体系.F1代优株快繁的最佳外植体为花葶和花蕾;消毒的最佳方案为用0.1%升汞进行消毒,处理时间为花蕾7.5 min,花葶8 min;诱导愈伤组织的最佳培养基为MS+NAA0.40 mg/L+6-BA2.0 mg/L,诱导率可达76.1%;最佳不定芽分化培养基为MS+6-BA2.0 mg/L+NAA0.30 mg/L,分化率可达94.03%;最佳生根培养基为1/2MS+IBA0.40 mg/L,生根率可达92.7%.The four excellent garden species adapted Harbin climate were chosen,which are the hybrids of Hemrocalis hybrida cv. ＂Zirong＂and H. fulva,and the rapid propagation system of the four F1superior groups was established in this paper. The best explants of F1are flower bud and scape. The best treatment is 0. 1% mercuric chloride and 7. 5min in flower bud and 8 min in scape. The most suitable medium inducing the callus,adventitious buds and root growing are MS ＋ NAA0. 40 mg / L ＋ 6-BA2. 0 mg / L,MS ＋ 6-BA2. 0 mg / L ＋ NAA0. 30 mg / Land1 /2MS ＋ IBA0. 40 mg / L,and their differentiation and rooting rates are 76. 1%,94. 03% and 92. 7% separately.
