摘  要:目的 观察缺氧复合梭曼中毒后大鼠肾上腺嗜铬细胞瘤PC12 细胞内游离钙的浓度 ([Ca2 + ] )、cAMP、钙调蛋白 (CaM)和钙调蛋白激酶Ⅱ (Ca2 + /CaM PKⅡ )的变化。方法 采用放免技术 ,从细胞水平进一步观察缺氧复合梭曼中毒对PC12 细胞的毒性、细胞内 [Ca2 + ]变化、cAMP、CaM和Ca2 + /CaM PKⅡ的变化。结果 缺氧中毒组PC12 细胞cAMP、CaM含量在中毒后 2 4h明显高于单纯中毒组、缺氧对照组和正常对照组 ;缺氧中毒组PC12 细胞Ca2 + /CaM PKⅡ活性在中毒后 2 4h明显低于单纯中毒组、缺氧对照组和正常对照组。结论  [Ca2 + ]、cAMP、CaM和Ca2 + /CaM PKⅡ在缺氧复合梭曼中毒PC12 细胞损伤机制中起了重要的作用。Objective To observe the changes of the concentration of Ca 2+ , contents of cAMP, CaM and activity of Ca 2+ /CaM PK II in pheochromocytoma PC 12 cells after combined soman and hypoxia injury. Methods The changes of [Ca 2+ ], and activity of CaM, cAMP and Ca 2+ /CaM PK II in PC 12 cells were studied after combined soman and hypoxia injury with radioimmunoassay. Results The changes of [Ca 2+ ], the contents of CaM, cAMP were significantly higher in hypoxic and soman intoxicated group than in soman intoxicated group and control group under hypoxia; but the activity of Ca 2+ /CaM PK Ⅱ were significantly decreased. Conclusion [Ca 2+ ], CaM, cAMP and Ca 2+ /CaM PK Ⅱ exert important role in the damage of PC 12 after combined soman and hypoxia injury.
