中药复方病毒克对兔HCLV模型组织病毒含量影晌的研究

摘  要:探讨抗病毒中药复方制剂的抗病毒作用机理,为提高临床疗效和扩大临床应用范围提供试验依据,本研究采用水萃取法制备抗病毒中药复方流浸膏制剂,以HCLV"C"株为模式病毒,GAPDH、ACTB为内参基因,以FQ-PCR作为检测方法,研究该中药复方制剂在对兔感染HCLV后脾脏、肠系膜淋巴结病毒增殖的变化情况。研究结果表明,高、中、低3个中药剂量组在连续5次给药后的24 h,即攻毒后的72、96、120、144 h,脾脏和肠系膜淋巴结中的HCLV含量低于对照组,脾脏中144 h病毒含量尤为明显(P<0.05),肠系膜淋巴结中72 h病毒含量尤为明显(P<0.05),说明该中药复方制剂在大剂量连续使用的情况下能明显抑制猪瘟兔化弱毒"C"株在淋巴结和脾脏中的增殖作用,并预示着这种抑制作用可能存在量效关系和时效关系。In order to provide experimental data for extensive clinical application, this study discussed the antiviral mechanism of traditional Chinese medicine compound preparation. The study adopted water extraction to make antiviral Chinese medicine Compound extractum, Taking the Hog cholera lapinized virus "C strain" as the modal virus, GAPDH and ACTB as reference gene, fluorescence quantitative PCR as testing method, adjusted value of Hog cholera lapinized virus as measurement indicators, the authors studied the effect of Chinese Herbal Compound on the propagation of virus in spleen and mesenteric lymph node after the rabbits were infected with HCLV. The study results indicate : 24 h after high-, medium- and low-dose groups of Chinese traditional medicine were given 5 times in a row, i.e. 72 h, 96 h, 120 h and 144 h after infection, HCLV content was lower than the control group in spleen and mesenteric lymph nodes, the virus content was particularly obvious at 144 h in spleen (P〈 0.05)and was particularly obvious at 72 h in mesenteric lymph nodes(P 〈 0,05). The results show that traditional Chinese medicine compound preparation can obviously inhibit the viral proliferative effect in the lymph nodes and spleen in the case of continuous use of high-dose drug, and indicate this inhibitory action may have dose-and time-dependent effects.
