摘  要:启动子是基因的重要组成部分,决定mRNA的转录,从而实现生物的有序发育过程。基于家蚕基因组和全长cDNA序列数据,鉴定了1 341条家蚕启动子序列。序列分析显示,在启动子-500~200 bp区间的GC、AT含量分别出现了1次和2次高峰值。TATA框(TATA-box)、起始子(Inr)、CAAT框(CAAT-box)和GC框(GC-box)等元件分别在256、453、200、334个启动子中被鉴定出,前3种元件集中分布于启动子-500~200 bp区间,第4种元件主要分布在-1 600^-300 bp上游较远处。进一步利用家蚕芯片表达谱数据发现23个启动子为精巢偏向表达基因的启动子。顺式元件分析这些精巢偏向表达启动子中含有6个高频率基序(motif),其中3个motif在精巢偏向表达启动子中的出现频率显著高于对照组,暗示这3个motif在精巢偏向表达基因的转录调控中可能起着重要作用。Promoter is an important component of genes that determines the transcription of mRNA to achieve the or- derly development and differentiation of organisms. In present study, we identified 1 341 promoters in silkworm ( Bom- byx mori) genomic and full-length cDNA sequence databases. Sequence analysis showed that GC content and AT content had one and two peak values in the promoter region from -500 to 200 bp, respectively. Elements of TATA-box, Initiator ( Inr), CAAT-box and GC-box were present in 256, 453, 200 and 334 promoters, respectively. The first three elements were dominantly located in the region from -500 to 200 bp, and the last element was mostly located at the upstream far from -1 600 to -300 bp. Silkworm-based genomic microarray data showed that 23 promoters are for tes- tis-biased gene expression. Cis-element analysis indicated that these promoters for testis-biased expression had 6 pre-dominant motifs, among which 3 motifs had significantly higher occurrence than the promoters for non-tissue specific expression, suggesting that these 3 motifs play an important role in the regulation of testis-biased gene transcription.
