摘 要:目的:按6.82 mg/(kg·bw)大鼠单剂量口服臭豆碱,用反相离子对高效液相色谱方法,测定大鼠肾组织中不同时间点臭豆碱的浓度。方法:高效液相色谱法,色谱柱为大连产HYP-ODS柱(200 mm×4.6 mm,5μm),柱温30℃;流动相由水相-异丙醇-乙腈(150∶15∶20体积比)组成。水相部分每1000 mL中含SDS 0.5g,三乙胺3.5 mL,85%磷酸2 mL(用三乙胺调至PH 3.0);流速1.0 mL·min-1紫外检测波长309 nm。结果:经DASver1.0药代动力学软件处理数据,臭豆碱在肾组织符合一室模型,其动力学参数为:Tmax(60)min,Cmax(13747.80)μg/L,T1/2(113.34)min,T1/2ka(16.19)min,AUC(3033978)μg/L·min,Vd/F(16.193 L/kg),CL/F(0.005)L/min/kg。结论:臭豆碱灌服后,可迅速分布到肾脏,具有消除快、蓄积时间短的特点。Objective : To measure the concentration of Anagyrine in its liver at different times by the way of an ion - pair liquid chromatographic (IPLC) by giving the rat a dose of 6.82 mg/( kg · b · w) orally. Method: The liquid chrom- atographic analysis was performed on a (LC - 10A) HPLC system (SHIMADZU), consisting of( LC - 10AT) pump. Separations were accomplished with a HYP -ODS C18 (200 mm x4.6 mm,5 μparticle size) from Dalian (China) ,op- erating at 30℃. The mobile phase consisted of water phase - isopropanol - acetonitrile ( 150 : 15 : 20) (v/v). Every 1000 mL of water phase contained 0.5 g of SDS ,3.5 mL of triethylamine and 2 mLof 85% phosphoric acid ( pH of mobile phase adjusted to 3.0 by addition of triethylamine). The mobile phase was filtered before using 0.45 μm nitrocellulose membrane and the flow rate was 1.0 mL · min-1 The UV detection wavelength was 309 nm. The detection limits of anagyrine was 10 μg · L^ - 1 ( S/N = 3 ). Results : The statistics was processed by a software dealing with medication dy- namics named DASverl. 0. It was found that the persistence pattern of Anagyrine in the liver was in agreement with the one - chamber model and its dynamic parameters were Tmax ( 60 ) rain, Cmax ( 13747.80 ) ) ng/g, TI/2 ( 113.34 ) min, T1/2k. ( 16.19)min, AUC(3033978) ng/( g · min), Vd/F ( 16. 193L/kg), CL/v (0. 005 ) L/( min· kg). Conclusion: After a dose of Anagyrine is given,it diffuses through the blood vessels into the kidney Immediately. It tends to vanish quickly without any remnant.
