采用UUO模型观察7种中药复方对慢性肾衰大鼠的疗效

摘　　要：目的:用7种不同治则的中药复方治疗UUO模型大鼠,观察这7种中药复方对慢性肾衰大鼠的治疗效果。方法:健康雄性SPF级SD大鼠100只,按体重随机分为10组,分别为:假手术组、模型组、健脾清化方组、肾衰三号方组、葆肾三号方组、慢性肾功能衰竭方组、滋补肝肾和化湿汤组、人参白术汤组、肾衰冲剂组、科素亚组,每组10只,用UUO的方法建立模型,灌胃14 d后处死,检测各组大鼠的血肌酐(Scr)、尿素氮(BUN)、24 h尿蛋白定量、尿β2微球蛋白(β2-MG)、尿N-乙酰-β-氨基葡萄糖苷酶(NAG)、尿转铁蛋白(TRF)的含量,评价不同治则对慢性肾衰大鼠的作用效果。结果:除假手术组外,各组大鼠肾功能出现失代偿,24 h尿蛋白定量、尿β2-MG、尿NAG、尿TRF的表达水平均有不同程度的增高;①与假手术组相比,模型组大鼠肌酐、尿素氮、24 h尿蛋白定量、尿β2-MG、尿NAG、尿TRF的表达水平均显著增高(P<0.01);②与模型组相比,各中药复方组大鼠肌酐、尿素氮、24 h尿蛋白定量尿β2-MG、尿NAG、尿TRF的表达水平均有不同程度的下降,各组相比较,以健脾清化方组下降程度最高(P<0.01)。结论:七种中药复方均能不同程度的改善慢性肾衰大鼠的肾功能,降低24h蛋白尿、尿β2-MG、尿NAG和尿TRF的表达,综合来说,健脾清化方作用更为突出。Objective ： To observe the effect of seven kinds of TCM compounds on chronic renal failure rats. Methods ： 100 healthy male SPF SD rats, according to the weight, were randomly divided into 10 groups, respectively is ：control group, model group, Jianpi Qianghua Fang group, Shenshuai Sanhaofang group, Baoshen Sanhaofang group, Manxing Shen- gongneng Shuaijie Fang group, Zibuganshen Huashi Decoction group, Renshen Baizhu Decoction group, Shenshuai Granule group,Cozaar group,each group of 10. Only with UUO method to establish the model, 14 days after gavage the rats were executed. The creatinine （ Scr）, urea nitrogen （ BUN）, 24 h urinary protein, urinary quantitative beta 2 microglobulin （beta 2- MG）,urinary n- acetyl- beta- d-glucosa minidase （NAG） and the content of urinary transferrin （agency） were detected. Results： In addition to the control group,each group of rats renal had been decompensation,24 hours uri- nary protein quantitative, beta 2 - MG urine, urine NAG and urine agency expression level were increased to some extent. （1）Compared with control group, model group rats＇ creatinine, urea nitrogen,24 h urinary protein quantitative, beta 2 - MG urine, urine NAG and urine agency expression levels were significantly increased （P 〈 0.01 ）. （2）Compared with model group, the Chinese medicine compound preparation group rats＇ creatinine,urea nitrogen,24 h urinary protein quantitative beta 2 - MG urine, urine NAG and urine agency the expression level all had different degrees of decline, of all group, Jian- pi Qinghua Fang group decreased the most （P 〈 0.01 ）. Conclusion： Seven kinds of Chinese medicine compounds can im- prove the renal function of chronic renal failure rats in different levels, reduce the 24 h proteinuria, beta 2 - MG urine, u- rine NAG and urine TRF, in aggregate, the effect of Jianpi Qinghua Fang is more outstanding.
