摘 要:目的:通过高脂诱导营养性肥胖大鼠模型,观察调脂积冲剂对肥胖大鼠体重、Lee’s指数、血清胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、血糖(FBG)、真胰岛素(FTI)、胰岛素抵抗指数(HOMA-IR)及游离脂肪酸(FFA)、脂联素、抵抗素的影响。方法:将SD雄性大鼠65只,分为正常组10只,其余55只采用高脂饮食建立肥胖模型。7周末时测体重,大鼠体重大于正常组体重20%为造模成功。将造模成功的30只大鼠随机分为模型组、调脂积组、荷丹片组。灌胃给药5周后,测量各组大鼠体质量、身长,计算Lee’s指数。检测各组大鼠血清TC、TG、HDL、LDL、FBG、FFA。采用ELISA法测定血清的真胰岛素、脂联素、抵抗素含量。采用稳态模式评估法(HOMA)来评价胰岛B细胞功能,即胰岛素抵抗指数(HOMA-IR)=(FBG)*FTI/22.5。结果:和正常组比较,模型组大鼠体重、Lee’s指数、TC、TG、LDL、FTI、HOMA-IR、FFA、抵抗素明显升高(P<0.01);FBG有差异性升高(P<0.05);HDL有差异性降低(P<0.05);脂联素明显降低(P<0.01)。和模型组比较,调脂积组大鼠体重明显下降(P<0.01);Lee’s指数、TG有差异性下降(P<0.05);TC、LDL明显下降(P<0.01);FBG略有下降、HDL略有升高,但无统计学意义(P>0.05);HOMA-IR、FTI、FFA明显下降(P<0.01);血清脂联素有差异性升高(P<0.05);血清抵抗素有差异性下降(P<0.05)。调脂积组和荷丹片组比较,在上述指标方面差异无统计学意义(P>0.05)。结论:高脂饲料诱导的肥胖大鼠体内存在胰岛素抵抗,血清FFA、抵抗素水平升高,脂联素水平降低;调脂积冲剂可以减轻肥胖大鼠的体重,调节血脂紊乱,改善其胰岛素敏感性,这种作用可能和降低血清FFA、抵抗素水平及升高脂联素水平有关。Objective: The obese rat model was induced by the high fat diet. To observe the influence of Tiaozhiji on weight of obese rat model, Lee's index, serum cholesterol ( TC), triglyceride ( TG), high density lipoprotein ( HDE), low density lipoprotein (LDL) ,blood glucose (FBG) ,true insulin ( FTI), insulin resistance index ( HOMA - IR), and free fatty acid ( FFA), adiponectin, resistin changes. To explore the mechanism of action of Tiaozhiji affecting body weight and insulin sensitivity in rat model. Methods: 65 male SD rats were divided into two groups, 10 of the normal group,the remai- ning 55 were made the obesity model by high fat diet. The body weight of rats was measured after 7 weeks. The weight 20% greater than the normal indicated modeling success. 30 successful modeling rats were randomly divided into model group,Tiaozhiji group and Hedan group. Gavage for 5 weeks,the rats body weight,length,calculation of Lee~ index were measured. Each group was detected the serum TC, TG, HDL - C and LDL - C, FBG and FFA. The enzyme - linked immu- nosorbent assay was used for determination of serum true insulin, adiponectin, resistin content. The homeostasis model as- sessment (HOMA) was used to assess the islet B cell function,insulin resistance index (HOMA -IR) = (FBG) × FTI/ 22. 5. Results : By comparison, the body weight of rats of model group was significantly increased than that in the normal group. Lee's index, TC, TG and LDL, FTI and HOMA -IR, FFA and resistin were significantly increased (P 〈 0. 01 ). FBG were increased in different degrees ( P 〈 0.05 ). HDL was decreased ( P 〈 0. 05 ). Comparison of model control group, the body weight of rats in the Tiaozhiji group was decreased significantly (P 〈 0.01 ). Lee's index and TG were de- creased significantly ( P 〈 0. 05). TC and LDL were decreased significantly (P 〈 0. 01 ). FBG was slightly decreased and HDL increased slightly but there was no statistical significance (
