宣肺止咳合剂抗小鼠肺组织损伤及NO水平的研究

摘　　要：目的：研究宣肺止咳合剂对内毒素所致急性肺损伤小鼠的保护作用机制。方法：取体重22～26g的健康清洁级雄性ICR小鼠50只，随机分为正常纽、模型纽、宣肺止咳舍剂高剂量（30mL／kg）、中剂量（15mL／kg）、低剂量（7．5mL／kg）组，每组10只。分组后，模型组给予蒸馏水灌胃，宣肺止咳合剂各剂量组以不同剂量的宣肺止咳合剂灌胃予以干预。灌胃给予宣肺合剂，1：k／d，连续7d，末次给药0．5h后，除正常组外所有小鼠腹腔注射脂多糖（LPS），在注射LPS后12h处死小鼠。以小鼠一般状态、肺组织含水量、血清NO水平、肺脏病理切片等指标评判宣肺止咳舍剂的保护作用。结果：各剂量组宣肺止咳合剂对急性肺损伤小鼠的一般状态均有一定的改善作用，并能抑制LPS导致的肺组织水肿，降低血清NO水平以及减少肺泡间质炎症细胞浸润。结论：宣肺止咳合剂对急性肺损伤小鼠有一定的保护作用。Objective：To observe the protective effects of Xuanfei Cough Mixture on acute lung injury in mice induced by endotoxin. Method： 50 healthy male ICR mice were randomized into normal group, model group, Xuanfei Cough Mixture -treatment group （high dosage, 30 mlml,/kg）, Xuanfei Cough Mixture- treatment group （middle dosage, 15 mL/kg） and Xuanfei Cough Mixture - treatment group （ low dosage,7.5 mL/kg）. Each group had ten animals. The model group was interfered with intragastric administration of distilled water, whereas each dose group of Xuanfei Cough Mixture was interfered with intragastric administration of Xuanfei Cough Mixture with different doses. 0.5 hour after the last administra- tion, and all mice were intraperitoneal injected lipopolysaccharide （LPS） to induce acute lung injury. 12 hours after LPS injection, l0 mice were sacrificed in each group, respectively. The general state of mice ,water content of lung tissue, ser- um level of NO and the lung biopsy were examined to judge the protective effect of Xuanfei Cough Mixture. Results ： Xuan- fei Cough Mixture, three doses groups, had some improvements on the general state of acute lung injury. Furthermore, the Xuanfei Cough Mixture inhibited the increase of lung wet weight and the serum level of NO, which caused by acute lung injury. In particular,it could relieve the progress of the alveolar interstitial inflammatory cell infiltration in mice. Conclu- sion：Xuanfei Cough Mixture has protective effect on acute lung injury in mice.
