甘草酸二铵胶囊及脂质复合物泡腾颗粒的小鼠体内分布

摘  要:目的:比较甘草酸二铵胶囊及其脂质复合物泡腾颗粒在小鼠体内的血和肝组织分布,探讨甘草酸二铵脂质复合物作为肝靶向药物的可行性。方法:以小鼠为实验动物,单次灌胃给予甘草酸二铵胶囊及其脂质复合物泡腾颗粒剂,剂量500 mg·kg-1,给药后30 min,1 h,2 h,4 h时取血浆及肝脏,高效液相色谱法测定血及肝脏中甘草酸二铵的浓度。结果:泡腾颗粒组小鼠血及肝组织中血药浓度显著高于胶囊组(P<0.01),肝组织中的药物浓度显著高于血中药物浓度(P<0.01),而胶囊组血及肝组织中血药浓度无统计学差异。结论:脂质复合物泡腾颗粒剂能显著提高小鼠血及肝组织中甘草酸二铵的浓度,特别是肝组织中药物的浓度,有肝靶向作用。Objective:To study the blood and liver tissue distribution of diammonium glycyn'hizinate (DG) capsules and its phosphatidylcholine complex (DGPC) effervescent granules in nfice and investigate the liver targeting effect of DGPC. Methods : The concentrations of DG in plasma and liver of mice were determined by HPLC after intragastric admin- istration with a dose of 500 mg· kg^- 1 of capsules and effervescent granules at 30 rain, 1 h, 2 h, 4 h. Results : The plasma concentration of DGPC effervescent granules group was significantly higher than that of capsule group ( P 〈 0.01 ), and liver tissue's drug concentration was significantly higher than drug concentration in plasma ( P 〈 0.01 ). However, there was no significant difference of drug concentration in plasma and liver tissue of mice in capsule croup. Conclusion:DGPC effervescent granule can significantly improve the concentration of Diammonium Glycyrrhizinate in plasma and liver tissue in mice, especially the drug concentration in liver tissue which has liver targeting.
