摘 要:目的:研究养血活血方对便秘老年小鼠肠神经递质的影响,阐明养血活血方治疗老年性便秘的作用机理。方法:选取健康老年小鼠,采用吗啡复制便秘小鼠模型,造模成功后,将60只健康老年ICR小鼠随机分为6组:空白组、模型组、阳性对照组(麻仁软胶囊)、养血活血方高、中、低剂量组。各组分别给药,连续灌服3周后处死,取结肠组织。采用免疫组化方法测定肠神经递质SP、VIP、5-HT的表达。结果:与空白组、阳性对照组及养血活血方各剂量组相比,模型组肠壁组织内SP、VIP、5-HT阳性表达明显减少,差异均有显著性统计学意义(P<0.01);各给药组组间比较可见:养血活血方高、中剂量组的神经递质SP、VIP、5-HT含量高于养血活血方低剂量、阳性对照组,差异有统计学意义(P<0.05或P<0.01)。结论:养血活血方可能通过增加肠神经递质SP、VIP、5-HT阳性表达来起到通便的作用,并且养血活血方的通便作用呈一定的量效依赖关系。Objective : To study the effects of Yangxue Huoxue Recipe (YXHYR) on intestinal neurotransmitters in old rats with colonic constipation, and to clarify the mechanism of Yangxue Huoxue Recipe in relieving elder constipation. Methods:The animal model with colonic constipation was established by old rats from subcutaneous injection with Mor- phine. 60 old rats with colonic constipation (slow transit colon) were divided into 6 groups:controL group, untreated group,Maren Soft Capsules - treated group, treatment group with high dosage YXHYR ( H - YXHYR), treatment group with middle dosage YXHYR (M -YXHYR), and treatment group with low dosage YXHYR (L- YXHYR). Each group was administered. 3 weeks, and then the colon specimen was obtained. Intestinal neurotransmitters including substance P ( SP), Vasoactive intestinal peptide (VIP) and 5 - HT were detected by immunohistochemistry method. Results: The ex- pressions of SP, VIP and 5 - HT in the untreated group were lower than those of the other groups (P 〈 0.01 ). Compared with the Maren Soft Capsules - treated group and L - YXHYR group, the expressions of SP, VIP and 5 - HT in the H - YXHYR and M -YXHYR groups were increased obviously (P 〈 0.05 or P 〈 0. 01 ). Conclusion: Yangxue Huoxue Reci- pe could have a purge role, and this may be due to its effect of regulating the expressions of SP, VIP and5 - HT in the in- testinal neurotransmitters, which shows a dose -effect relationship.
