摘 要:目的:探讨辛黄汤对组胺诱导的离体变应性鼻炎(allergic rhinitis,AR)黏膜组织的作用机制。方法:用组胺诱导离体AR黏膜组织模型,然后分组给予布地奈德、辛黄汤高、中、低剂量培养;用HE染色病理学观察不同浓度的辛黄汤对离体鼻黏膜组织的病理学效用;用FCM检测不同浓度的辛黄汤对组胺诱导的离体鼻黏膜组织细胞以及ANI或zVAD-fmk预孵0.5 h后的组胺诱导的离体鼻黏膜组织细胞的凋亡率;用Western blot检测凋亡相关蛋白的表达情况。结果:不同浓度的辛黄汤作用于组胺诱导的离体AR鼻黏膜组织24 h后,损伤的鼻黏膜组织结构渐趋完整,层次逐渐清晰,水肿明显减轻,呈浓度依赖性;鼻黏膜组织细胞的凋亡率渐趋减少,凋亡率呈浓度依赖性(P<0.05);同时凋亡相关蛋白cyto-c、Caspase-3和PARP蛋白表达降低(P<0.05),而经ANI或zVAD-fmk预孵0.5 h后组胺诱导的离体鼻黏膜组织细胞的凋亡率无明显变化(P>0.05)。结论:辛黄汤可以显著改善组胺诱导的离体AR黏膜组织病理学症状,其机制可能与其调节内源性线粒体凋亡途径相关。Objective : The aim of this study is to investigate the effect of Xinghuang Decoction on histamine - induced nasal tissue of allergic rhinitis and Its Possible Mechanism in vitro. Methods:The models of histamine -induced nasal tis- sue of allergic rhinitis were made in vitro, and exposed to Budesonide and various concentrations of Xinghuang Decoction for 24 h respectively, liE staining pathological observation was used to evaluate the effects of the above experimental med- icines. The apoptotic rates of the histamine - induced nasal tissue of allergic rhinitis received the experimental medicines or treated with ANI or zVAD - fmk half an hour earlier and then received the experimental medicine were detected by flow cytometry (FCM) , and the protein expressions related to apoptosis were examined by western blot. Results:The damaged structure of histamine - induced nasal tissue of allergic rhinitis was coming to recover and the anatomical level turned to clear gradually. The edema was significantly relieved in a dose - dependent manner. The apoptotic rate of nasal tissue of allergic rhinitis was gradually decreased (P 〈 0.05 ), and the protein expressions of cyto-c, Caspase - 3 and PARP were down regulated ( P 〈 0.05 ) when they received different concentrations of Xinhang Decoction. While the apoptotic rate of histamine - induced nasal tissue of allergic rhinitis which was treated with ANI or zVAD - fmk half an hour earlier had not altered ( P 〉 0. 05 ). Conclutions : Xinhuang Decoction could significantly alter pathological symptoms of histamine - in- duced nasal tissue of allergic rhinitis,which may be associated with mitochondiral apoptotic pathway.
