摘  要:目的观察中药肾络通对梗阻性肾病大鼠氧化应激和细胞凋亡的影响及可能作用机制。方法 48只Wistar大鼠随机分为假手术组、模型组、Tempol组和肾络通组,每组12只。除假手术组外,其余各组大鼠结扎左侧输尿管建立梗阻性肾病动物模型,造模后Tempol组以Tempol 30mg/(kg·d)入水瓶中饮用;肾络通组予肾络通煎剂26g/(kg·d)饮用,持续10天。HE、Masson染色观察各组大鼠梗阻侧肾脏组织形态学改变;酶联免疫吸附法(ELISA)测定血清、尿液8-羟基脱氧鸟苷(8-OHdG),比色法测定血清丙二醛(MDA)含量;TUNEL法及光镜下观察肾组织细胞凋亡;免疫组织化学法检测大鼠肾组织半胱氨酸-天冬氨酸蛋白水解酶-9(Caspase 9)及半胱氨酸-天冬氨酸蛋白水解酶-12(Caspase 12)蛋白表达。结果与假手术组比较,模型组大鼠肾间质大量炎性细胞浸润,胶原沉积显著增加;血清、尿液8-OHdG和血清MDA含量显著升高(P<0.05或P<0.01);细胞凋亡阳性细胞显著增多(P<0.01);Caspase 9、Caspase 12蛋白表达明显上调(P<0.01)。与模型组比较,Tempol组及肾络通组炎性细胞浸润及胶原沉积均减少,血清、尿液8-OHdG和血清MDA含量显著降低(P<0.05或P<0.01);细胞凋亡阳性细胞显著减少(P<0.05或P<0.01);Caspase 9、Caspase 12蛋白表达明显下调(P<0.05)。结论中药肾络通可减轻梗阻造成的氧化损伤,提高抗氧化能力,降低脂质过氧化水平,抑制细胞凋亡,其机制可能与降低Caspase 9、Caspase 12蛋白有关,并且效果与抗氧化剂Tempol相当。ABSTRACT Objective To research the effects and mechanisms of Shenluotong ('~) on oxidative stress and apoptosis in rats with obstructive nephropathy. Methods Totally 48 female Wistar rats were randomized into the sham operation group, model group, Tempol group and Shenluotong (肾络通) group, with 12 in each. The rat models were established by ligation of the left ureter in all groups except the sham operation group. The rats in the Tempol group and Shenluotong (肾络通) group were fed with 30 mg/ ( kg . d) Tempol and 26 g/ ( kg .d) Shenluotong (肾络通) respectively for 10 days. The morphological changes of obstructed kidneys were observed by HE staining and Masson staining. Enzyme-linked immunosorbent assay (ELISA) was used to detect serum and urine 8-hydroxy-deox- yguanosine (8-OHdG). The content of malondialdehyde (MDA) was detected by colorimetric assay. The apoptosis was observed by TUNEL assay and light microscope. The protein expressions of Caspase 9 and Caspase 12 were detec- ted by immunohistochemistry and Western blot. Results Comparing with the sham operation group, a large number of inflammatory cells were infiltrated and collagen deposition was increased in renal interstitium, the contents of 8 OHdG and MDA were significantly increased (P 〈 0. 01 or P 〈 0.05 ), positive apoptotic cells were significantly in creased ( P 〈0. 01 ) and the protein expressions of Caspase 9 and Caspase 12 were significantly increased in the model group (P 〈 0. 01 ). Comparing with the model group, the infiltration of inflammatory cells and collagen deposition was decreased, the contents of 8-OHdG and MDA were significantly decreased (P 〈0.01 or P 〈0. 05 ) , positive apoptotic cells were significantly decreased (P 〈0.0l or P 〈 0.05 ) and the protein expressions of Caspase 9 and Caspase 12 were significantly decreased in the Tempol group and Shenluotong (肾络通) group (P 〈0.01 ).
