复方鳖甲软肝片配伍对芍药苷在小肠吸收的影响

摘　　要：目的研究复方鳖甲软肝片在模拟小肠环境中各成分的吸收情况,考察复方配伍对其指标成分芍药苷吸收的影响,为选择复方鳖甲软肝片的质量控制指标提供参考。方法通过外翻肠囊法模拟复方鳖甲软肝片在小肠的吸收过程,收集3种浓度的复方鳖甲软肝片给药后与芍药苷单独给药后不同时间的肠囊液样品,用HPLC检测样品中的芍药苷、连翘苷、人参皂苷Rg1,绘制时间-累积吸收曲线,比较复方与单用芍药苷吸收量的差异。结果复方鳖甲软肝片不同浓度时各成分在不同肠段的吸收均为线性吸收,其回归方程的相关系数(r2)均大于0.9,符合零级吸收;其吸收速率常数Ka随药物浓度的增加而增大,说明其为被动吸收。与单成分相比,芍药苷在复方中的累积吸收率较好。结论复方鳖甲软肝片的药物配伍有利于芍药苷吸收,人参皂苷Rg1可增加作为复方鳖甲软肝片质量控制指标。OBJECTIVE To study the intestinal absorption characteristics of three components （ paeoniflorin, phillyrin, ginsen- osides Rgl ） of Fufangbiejiaruangan tablets and the effects of prescription on the intestinal absorption of paeoniflorin. METHODS E verted intestinal sac method was adopted to mimic the intestinal absorption process of Fufangbiejiaruangan tablets extract at three con centrations. The intestinal absorption mechanisms of paeoniflorin, phillyrin and ginsenoside Rgl were studied. HPLC assay was used to determine the three components and the time-accumulative absorption curves were drawn to compare the absorption characteristics of paeoniflorin in the prescription preparation and paeoniflorin solution. RESULTS The absorption of the three components was linear at different intestinal segments and doses, and the correlation coefficients of the regression equations were all more than 0. 9 （ r^2 〉 O. 9 ） , which were consistent with zero order rate process. The Ka（s） of the components increased along with the increasing dosage, which in dicated passive absorption. The cumulative absorption rate of paeoniflorin in Fufangbiejiaruangan tablets was better than one-component solution. CONCLUSION The compatibility of Fufangbiejiaruangan tablets is beneficial to paeoniflorin absorption and ginsenoside Rgl may be the indicator of the quality standard of Fufangbiejiaruangan tablets.
