摘 要:在pH6.5的Walpole缓冲溶液中,当以300nm的光激发时,1,2-萘醌-4-磺酸钠在468nm处产生1个较强的荧光峰。加入异烟肼后,由于其与1,2-萘醌-4-磺酸钠发生反应,从而使体系颜色加深且荧光信号减弱。在最佳实验条件下,在0.10—8.0μg/mL范围内,体系的荧光信号△F与异烟肼浓度之间有较好的线性关系,检出限为40.2ng/mL。方法可直接用于测定药物中异烟肼含量,回收率为96.5%~106.3%。Sodium 1,2-naphthoquinone-4-sulfonate produced a strong fluorescence peak at 468 nm in the Walpole buffer solution of pH 6.5 at an excitation wavelength of 300 nm. The color of the system is deepened and the fluorescence signal is weakened with addition of isoniazid because of isoniazid reacting with sodium 1,2- naphthoquinone-4-sulfonate. Under the optimum experimental conditions, there is a good linear relationship between the fluorescence signal and the concentration of isoniazid in the range of 0.10 - 8, 0 μg/mL with the detection limit of 40.2 ng/mL. This method can be directly used for the determination of isoniazid content with the recoveries of 96.5% - 106.3%.
