摘 要:目的观察清肠化湿方对实验性大鼠结肠炎结肠黏膜ICAM-1表达的影响,以探讨其治疗溃疡性结肠炎(UC)的可能机制。方法使用TNBS/无水乙醇造模成功后,分为空白组、模型组、清肠化湿方组、美沙拉嗪组。连续灌胃给药10d后,处死大鼠,留取结肠组织,免疫组化法及ELISA法观察结肠组织ICAM-1的表达。结果清肠化湿方可以显著减少大鼠结肠炎模型结肠粘膜ICAM-1的表达。结论清肠化湿方可以减少结肠粘膜ICAM-1的表达,从而抑制白细胞的迁移和活化,是其治疗UC的机制之一。Objective To explore the possible mechanisms of the Qingchang Huashi decoction's therapeutic effect on Ulcerative Colitis(UC) ,through observing the expressive levels of ICAM - 1 in colonic mucosa. Methods We use TNBS/anhydrous alcohol to make model. Then, model rats were divided into model,Qingchang Huashi and Mesalazine group. And a group of rats untreated was used as blank group. Qingchang Huashi and Mesalazine group rats were irrigated to stomach with drug dilutions for 10 days successively. Then, all groups of rats were killed and colon tissues were cut out for immunohistoehemical and ELISA inspections. Results The ICAM - 1 expressive levels enhanced in model rats. The Qingchang Huashi decoction can decrease the ICAM - 1 (P 〈 0.01 ). The curative effect between QingchangHuashi and Mesalazine group has no significant difference(P 〉 0.05 ). Conclu- sion Qingcbang Huashi decoction can decrease the ICAM - 1 in colonic mucosa,which is the one of the mechanisms to treat UC.
