赤雹根总皂苷对佐剂型关节炎大鼠的抗炎作用及TNF-α,IL-1β和IL-6mRNA表达的影响

摘　　要：目的观察赤雹根总皂苷(STDR)对大鼠佐剂型关节炎(AA)的治疗作用及对其炎症组织中TNF-α,IL-1β和IL-6 mRNA表达水平的影响,探讨STDR治疗类风湿性关节炎的作用机制,为临床应用提供实验依据。方法雄性SD大鼠60只,随机分为正常对照组、模型组、STDR 40mg·kg-1·d-1,80mg·kg-1·d-1,160mg·kg-1·d-1剂量组和雷公藤多苷12mg·kg-1·d-1对照组。除正常对照组外,其余各组大鼠均于右后足跖部皮下注射完全弗氏佐剂0.1ml。造模18天后各给药组均ig给予相应药物,模型对照组ig给予等体积蒸馏水,给药21 d。于给药后第21天处死大鼠,取其足跖部,采用RT-PCR法提取炎症组织中mRNA测定TNF-α、IL-1β和IL-6 mRNA的表达水平,并取膝关节滑膜组织进行病理学观察。结果 TNF-α、IL-1β和IL-6 mRNA的表达水平明显著低于模型组(P<0.05或P<0.01),膝关节滑膜增生及炎症反应明显轻于模型组。结论 STDR对大鼠AA具有显著的治疗作用,其抑制TNF-α、IL-1β和IL-6mRNA的表达可能是其治疗AA的机制之一。Objective To study the effect of total saponins of Thladiantha dubia root on the joint swelling degree and the expres- sion levels of TNF - α mRNA and IL - 1 β mRNA in adjuvant arthritis rats. Methods Male rats were divided in six groups of ten each. The normal control group, model group, groups of STDR 160mg·kg^-1·d^-1, 80mg·kg^-1·d^-1 , 40mg·kg^-1·d^-1 and group of Tripterygium Glycoside 12 mg·kg^-1·d^-1. Except normal control group rats, the remaining group rats were in the right foot plantar subcutaneous injection of FCA 0. lml. The STDR groups and Tripterygium Glycoside group rats intragastric ad- ministration for 21 days on the 18th days after injected FCA. The model control group ig received equal volume of distilled water. And the rats were killed at 21st days after administration , plantar subcutaneous were received . The expression of TNF - α mRNA, IL - 1β mRNA , IL - 6 mRNA was determined in the inflammatory tissue by RT - PCR. The pathology of vola pedis was ob- served. Results The expression of TNF - α, IL - 1 β, IL - 6 mRNA in STDR groups were lower than those in model group （ P 〈 0.01 ）. Conclusion The STDR has Anti - inflammatory effect on AA rats. The role is closely related to the inhibition on the ex- pression of the mRNA of cytokines TNF -α, IL - 1 β and IL - 6.
