复方“前愈”总生物碱部位的分离纯化及含量测定

摘　　要：目的从中药复方"前愈"中提取分离总生物碱化学部位并将其进行纯化,建立总生物碱及盐酸小檗碱的含量测定方法。方法采用溶剂法分离纯化总生物碱化学部位;应用紫外分光光度法和高效液相色谱法,分别测定分离提取液及纯化液中总生物碱及盐酸小檗碱的含量。结果溶剂法提取分离所得总生物碱含量为2.066%,盐酸小檗碱含量为1.59%;纯化后总生物碱含量为46.52%,盐酸小檗碱含量为13.98%。结论实验确定的分离纯化方法操作简便合理,可显著提高"前愈"复方中总生物碱及盐酸小檗碱的含量。Objective To extract and purify from traditional Chinese medicine ＂ Qianyu＂ of the total alkaloid chemical parts and make them purified, and to establishthe method for the determination of total alkaloids and berberine hydrochloride. Methods - Solvent separation and purification of total alkaloid chemical parts ; using UV spectrophotometry and high performance liquid chro- matography, were determined the content of total alkaloids and berberine hydroehloride separated extract and purified liquid. ]Re- sults The solvent extraction separation resulting total alkaloid content of 2. 066% , Berberine content of 1.59% ; purified total al- kaloid content of 46.52% , Berberine content of 13.98%. Conclusion The separation and purification method determined by this experiment is simple and reasonable, which can significantly improve ＂Qianyu＂ total alkaloids in the compound, and the content of berberine hydrochloride.
