艾灸对肝癌裸鼠移植瘤PCNA、CyclinD1、CDK4表达影响的实验研究

摘　　要：目的观察艾灸对肝癌裸鼠移植瘤增殖细胞核抗原(PCNA)、细胞周期蛋白D1(CyclinD1)和周期蛋白依赖性激酶4(CDK4)表达的影响,探讨艾灸抗肿瘤的可能机制。方法 16只裸鼠均接种HepG2细胞构建高转移肝癌小鼠移植瘤模型,随机分为模型组和艾灸组。接种后次日给予麦粒灸大椎穴干预,至14 d后结束治疗,取出肿瘤组织。采用免疫组化法检测肿瘤组织中PCNA蛋白的表达,PCR法检测CyclinD1、CDK4mRNA的表达。结果与模型组相比,艾灸组中PCNA蛋白、CyclinD1mRNA和CDK4mRNA表达均显著降低(P<0.05)。结论艾灸对肝癌裸鼠移植瘤的生长有明显的抑制作用,其机制可能与下调肿瘤组织中PCNA、CyclinD1mRNA、CDK4mRNA的表达水平有关。Objective To observe the efficacy of PCNA, CyclinD1 and CDK4 in the expression of hepatocellular carcinoma in nude mice treat with moxibustion and explore the possible mechanism of antitumor through moxibustion. Methods 16 nude mice were inoculated with HepG2 cells to construct high metastatic hepatocellular carcinoma xenograft model in mice and randomly divided into model group and moxibustion group. The next day is to be intervened with DU 14 with Moxibustion. After treatment of 14 days, the tumor tissue in nude mice were removed. Using immunohistochemical method to detect the expression of PCNA protein and PCR method to detect the expression of CyclinD1 and CDK4mRNA in tumor tissues. Results Compared with the model group, the expression of PCNA CyclinD1 and CDK4mRNA were significantly lower（ P 〈 0.05 ）. Conclusion Moxibustion has significantly inhibited on the growth of transplanted tumor and the mechanism may be related with down - regulation of the expression level of PCNA CyclinDlmRNA, CDK4mRNA in tumor tissues.
