摘  要:目的:基于中医"肝肾同源"理论,观察中药方剂加味消渴康对糖尿病肾病(diabetic nephropahy,DN)大鼠肝组织转化生长因子-β1(transforming growth factor-β1,TGF-β1)、Smad-4蛋白(drosophila mothers against decapentaplegic protein-4)、α-平滑肌肌动蛋白(alpha smooth muscle actin,a-SMA)和血小板衍生生长因子(platelet-derived growth factor,PDGF)表达的影响。方法:采用单侧肾切除加链脲佐菌素(streptozotocin,STZ)诱导的糖尿病肾病大鼠模型,将造模成功后的DN大鼠按随机数字表法分为正常组、模型组、西药组和中药小、中、大剂量组。其中西药组给予氯沙坦灌胃,中药组给予加味消渴康灌胃,正常组及模型组每日灌服等量蒸馏水共12周。各组大鼠于第12周末处死取出肝脏,运用ELISA法观察大鼠肝组织TGFβ1、Smad-4、a-SMA和PDGF的表达。结果:与正常组比较,各组大鼠肝中TGFβ1、Smad-4、a-SMA和PDGF的表达明显升高,差异有统计学意义(P<0.01)。中药治疗后,各组大鼠TGFβ1、Smad-4、a-SMA和PDGF的表达有一定程度的降低,差异有统计学意义(P<0.01或P<0.05)。结论:加味消渴康对DN大鼠的肝损伤均有保护作用。Objective : To investigate the influence of chinese herbal medicine prescription "JiaweiXiaokekang" on the expression of TGF-β1 (transforming growth factor-β1 ) ,Smad-4(drosophila mothers against decapentaplegic protein-4) , α- SMA( alpha smooth muscle actin) and PDGF(platelet-derived growth factor) in the hepatic tissues of diabetic nephropathy (DN) rats based on the theory of "homogeny of liver and kidney". Methods : The model was established by uninephrectomy and injection of streptozotocin(STZ). Models were divided into normal group, model group, western medicine group and Chinese herb medicine group which includirg low-dose, medial-dose and high-dose group. Western medicine group was given losartan by intragastric administration; Chinese herb medicine group was given JiaweiXiaokekang by intragastric administration; The normal group and model group were fed with equal distilled water daily. After 12weeks, the rats in each group were sacrificed and removed the hepars and then observed the expressions of TGF-β1, Smad-4, a-SMA and PDGF by ELISA. Results:The expression of TGF-β1 ,Smad-4,α-SMA and PDGF in other group was higher than that in normal group, there was significant difference between them (P 〈 O. O1 ). After the treatment of Chinese herbs, their expression was lower, there was significant difference(P 〈 0. 01 or P 〈 0.05 ). C onclusion:JiaweiXiaokekang has a certain protective effect on the hepatic injury of DN rats.
