摘  要:目的:采用RNAi技术沉默LIMK2基因来抑制细胞骨架改建,观察其对流体剪切力下成骨细胞COX-2基因力学敏感性的影响。方法;RNAi技术沉默原代小鼠成骨细胞LIMK2基因来抑制细胞骨架改建,分别用实时荧光定量巢式PCR和免疫荧光的方法检测流体剪切力下COX-2基因mRNA和蛋白的表达水平。结果:RNAi抑制细胞骨架改建可以促进流体剪切力诱导的成骨细胞COX-2 mRNA和蛋白的表达(P<0.05)。结论:采用RNAi技术抑制细胞骨架改建,可以提高流体剪切力作用下成骨细胞COX-2基因的力学敏感性。Objective: To study the effect of cytoskelet on reorganization in hibition with RNA interference on the mechanosensitivity of COX -2 gene in osteoblasts induced by fluid shear stress.Methods: RNA interference against LIMK2 gene was used to inhibit the cytoskeleton reorganization of BALB/cmouse prim ary osteoblasts.Osteoblasts were loaded by fluid shear stress at12 dyne/cm 2 or unloaded.The nested real-time PCR and immun of luorescence were performed to detect the expression levels of COX-2m RNA and protein, respectively. Result: Cytoskeleton reorganization inhibition with RNA interference against LIMK2 gene increased the expression of COX -2 gene induced by fluid shear stress.Conclusion: Cytoskeleton reorganization inhibition with RNA interference against LIMK2 gene could enhance the mechanosensitivity of COX -2 gene induced by fluid shearstress.
