摘 要:目的研究去辣椒素敏感性感觉神经对大鼠急性心肌缺血心脏Cu—Zn超氧化物岐化酶(SOD1)和谷胱甘肽S一转移酶-P(GST—P)表达的影响。方法将新生48hSD雄性乳鼠随机分为正常组和去辣椒素敏感性感觉神经组,去辣椒素敏感性感觉神经组用50mg·kg。。辣椒素皮下注射,正常组用溶剂皮下注射,待乳鼠复苏后置于鼠笼,饲养至250~290g造急性心肌缺血模型。各组再分为假手术组(正常假手术组、去辣椒素敏感性感觉神经假手术组)和急性心肌缺血组(正常心肌缺血组、去辣椒素敏感性感觉神经心肌缺血组)两个亚组。在缺血6h时各组大鼠均被处死,解剖取左室壁心肌组织,用于提取总蛋白,Westernblotting检测各组大鼠SOD1、GST—P蛋白质的相对表达量。结果与正常假手术组比较,正常心肌缺血组SOD1、GST—P表达显著降低(P〈0.05,P〈0.01);去辣椒素敏感性感觉神经假手术组SOD1表达显著降低(P〈0.01),GST—P表达差异无统计学意义(P〉0.05)。与去辣椒素敏感性感觉神经假手术组比较,去辣椒素敏感性感觉神经心肌缺血组SOD1、GST—P表达差异均无统计学意义(P〉0.05)。与正常心肌缺血组比较,去辣椒素敏感性感觉神经心肌缺血组SOD1表达显著降低(P〈0.05),GST—P表达差异无统计学意义(P〉0.05)。结论辣椒素敏感性感觉神经对急性心肌缺血心脏中SOD1、GST—P在一定程度上有调控作用。Objective To study the impacts of capsaicin -sensitive sensory nerves (CSSN)on the heart Cu - Zn superoxide dismutase( SOD1 ) and glutathione S transferase - P( GST - P) in the rats of acute myocardial ischemia. Methods The 48 h newly - born SD male rats were randomized into a normal group and a CSSN group. In CSSN group, capsaiein,50 mg ~ kg- l was used for subcutaneous injection. In the nor- mal group, the solvent subcutaneous injection was applied. After resuscitation, the suckling rats were placed in the cage and fed till weighted 250 - 290 g and the models of acute myocardial ischemia were prepared. Each group was subdivided into pseudo- operation subgroups (a normal pseudo- operation subgroup, a CSSN pseudo- operation subgroup)and acute myocardial ischemia subgroups( a normal myocardial ischemia sub- group, a CSSN myocardial ischemia subgroup). In 6h of ischemia, the rats in each subgroup were sacrificed. The myocardial tissue of the left ventricular wall was collected for total protein extraction. Western blotting method was used to detect the relative transcript level of SOD1 and GST - P proteins. Results Compared with the normal pseudo - operation subgroup, SOD1 and GST - P expressions were reduced significantly in the normal myocardial isehemia subgroup( P 〈 0.05, P 〈 0.01 ), SOD1 expression was reduced significantly in CSSN pseudo -operation subgroup (P 〈 0.01 ) ;the difference in GST- P expression was not significant( P 〉 0.05 ). Compared with CSSN pseudo- operation subgroup ,SOD1 and GST- P expressions were not differ- ent significantly in CSSN myocardial isehemia subgroup( P 〉 0.05 ). Compared with the normal myocardial is- chemia subgroup, SOD1 expression was reduced significantly in CSSN myocardial ischemia subgroup (P 〈 0.05 ) and GST - P expression was not different significantly (P 〉 0.05 ). Conclusion CSSN plays the regu- latory effect on heart SOD1 and GST- P in acute myocardial ischemia to a certain degree.
