摘 要:目的观察奥美拉唑体内外给药对壁细胞H+-K+-ATP酶活性的影响。方法体内给药:大鼠分为空白对照组、奥美拉唑组,奥美拉唑组体内给予奥美拉唑5 mg·kg-1,连续3 d,提取分离壁细胞,检测H+-K+-ATP酶活性;体外给药:分离纯化大鼠壁细胞,进行裂解后,随机分为空白对照组、奥美拉唑组,奥美拉唑组给予奥美拉唑4μg·mL-1,水浴孵育30 min,检测H+-K+-ATP酶活性。结果奥美拉唑体内给药未见明显抑制壁细胞H+-K+-ATP酶活性的作用,体外给药可明显抑制壁细胞H+-K+-ATP酶活性。结论分离纯化壁细胞后进行药物干预效果更明显,可作为药物筛选的方法之一。Objective To observe the impacts on parietal cell H+-K+-ATP enzymatic activity with the medication of Omeprazole(OME)in vitro and vivo. Methods Medication in vivo:the rats were di- vided into a blank control group and OME group. In OME group, OME was prescribed for 3 days continuous- ly ,5 mg, kg-l The parietal cells were extracted and separated for the determination of H + -K + -ATP en- zymatic activity. Medication in vitro : After the separation and purification of parietal cells in the rats as well as cell lysis, the rats were randomized into a blank control group and OME group. In OME group, OME was pre- scribed ,4 p^g ~ mL-1. After water bath for incubation for 30 rain, H + - K + - ATP enzymatic activity was de- termined. Results Medication in vivo with OME did not present the obvious suppression on H + - K + - ATP enzymatic activity. The medication in vitro indicated the apparent suppression on H+-K+-ATP enzymatic activity. Conclusion Medicinal intervention after the separation and purification of parietal cell indicates the apparent effect and it can be one of the methods for drug screening.
