摘  要:建立了高效液相色谱-二极管阵列检测器(HPLC-PDA)分离测定口含烟中尼古丁的方法。样品经过磷酸二氢盐水溶液(pH=9.0)提取后,以AgilcntZorbax-SBC18.5μm、4.6mm×250mm为色谱柱,10mmol/L磷酸二氢钠水溶液(含0.1%---乙胺,pH=3.O)和甲醇为流动相,200---500nm波长范围内进行扫描检测。结果表明,在8min以内尼古丁得到了较好的分离和检测,在O.5~100mg/L范围内具有较好的线性(,印.9997),检出限为0.18mg/L,3种不同浓度水平的加标回收率在89.7%-109.8%之间,相对标准偏差在6%以内。该方法操作简单。灵敏度高。可以满足口含烟中尼古丁的检测。A high performance liquid chromatography method coupled with Photo Diode Array Detector (PDA) as detector was established for the separation and determination of nicotine in snus samples. After extracted by phosphate salt aqueous solvent (pH=9.0), the target compound were separated on a Agilent Zorbax-SB Cls column (51am, 4.6mm×250mm), using HPLC methanol and 10mmol/L monosodium orthophosphate aqueous solution with 0.1% (V/V) triethylamine as mobile phases, and detected by a diode array detector scanned from 200 nm to 500nm. It's exhibited that there was a good linear relationship (r=0.9997) ranged from 0.5 to 100 mg/L and the limits of detection was 0.18mg/L. The average recoveries at three levels were in the range of 89.7%--109.8% with the relative standard deviations (RSD) less than 6%. This simple method can be used to determine nicotine in snus samples with good reproducibility.
