摘 要:为了研究高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)HuN4株在其致弱过程中对仔猪外周免疫器官及肺脏损伤的变化,试验分别采用HuN4F5及其致弱代次F15、F23、F30及HuN4弱毒疫苗F112感染30日龄PRRSV抗原抗体阴性健康断奶仔猪,每天进行体温测定及临床症状观察,并在感染后0,3,5,7,10天检测病毒血症情况。病毒感染后10天剖杀试验猪,观察外周免疫器官及肺脏的病理变化并对肺脏和主要外周免疫器官病毒载量进行测定。结果表明:HuN4 F5、HuN4 F15组在临床症状、病毒血症、外周病毒载量及病理变化方面都明显比其他组严重,HuN4 F23组发病明显减轻,HuN4 F30组、HuN4 F112组无明显发病特征,与空白对照组无明显差异。说明HP-PRRSV HuN4株在致弱过程中,传代至30代时毒力发生显著减弱。To study the lesion changes in peripheral immune organs and lung of the piglets infected with HP - PRRSV HuN4 in the course of its attenuation, the HuN4 strain (F5), its attenuated generations in F15, F23, F30, and the attenuated vaccine HuN4 F112 were used to infect the 30 - day - old healthy pigs tested negative for PRRSV antigen/antibody. All measurements of the body temperature were carried out and the clinical symptoms were observed every day, the HuN4 viremia was detected on days of 0, 3, 5,7,10 days after infection. The experimental pig- lets were sacrificed at 10 days after the virus infection, and then the pathological changes in peripheral immune organs and lung of the piglets were observed, and the viral loads of the major peripheral immune organs were measured. The results showed that the piglets of the HuN4 F5 and HuN4 F15 groups, were significantly wo^e than those of the other groups in terms of the clinical symptoms, viremia, viral loads and patho- logical changes in peripheral immune organs and lung. The clinical symptoms in the HuN4 F23 group were significantly reduced, and there were no obvious clinical symptoms between the HuN4 F30 and HuN4 F112 groups, which was no significant difference compare with the blank con- trol group. The results indicate when the HP - PRRSV HuN4 strain in the course of its attenuation was passaged for 30 generations, the viru- lence of the HuN4 F30 strain is significantly decreased.
