WT1基因在猪泌尿生殖器官中表达情况的研究

摘  要:为了研究维尔姆氏瘤基因1(WT1基因)在成体猪各器官表达水平的差异,以及WT1基因在猪胚胎期和成体两个阶段泌尿生殖器官的表达位置,试验以猪作为研究对象,利用荧光定量PCR和免疫荧光染色等技术,对WT1基因在胚胎时期和成体猪各器官的表达情况进行了研究。结果表明:WT1基因在成体猪心脏、脾脏、肾脏、卵巢和睾丸中高表达,在肝脏和肺脏中低表达;在猪胚胎时期主要表达在肾脏中的comma-小体和S-小体、卵巢的皮质和睾丸中的曲细精索周围的间质细胞中;在成体中WT1基因主要表达在肾小管、睾丸曲细精管周围的间质细胞和卵巢的卵泡膜外层细胞中。说明WT1基因对猪胚胎时期泌尿生殖器官的发育、形成以及成体泌尿生殖器官功能的维持发挥重要作用。To study the differences of the expression levels of the Wilms' tumor 1 (WT1) gene in various organs from adult pigs, as well as the positions of the expression of the WT1 gene in porcine genitourinary organs during embryonic and adult stages. The WT1 gene expressions in porcine genitourinary organs in the two stages were studied by using fluorescence quantitative PCR and immunofluorescenee staining techniques and so on with the pigs as research animals. The results showed that the WTI gene was highly expressed in heart, spleen, kidney, ovary and testis, and less expressed in liver and lung in adult pigs. The WT1 gene was mainly expressed in comma - and S - shaped bodies of the kid- ney, ovarian cortex and testicular interstitial cells surrounding seminiferous cords in embryonic pigs ; The WT1 gene was mainly expressed in re- nal tubules, testicular interstitial cells surrounding seminifereus tubules and the outer layer cells of ovarian follicle membrane in adult pigs. The results indicate that the WTI gene plays an important role in the development and formation in embryonic pigs, and functional maintenance of genitourinary organs in adult pigs.
