摘 要:为了研究蛇床子中蛇床子素的最佳提取工艺,试验以蛇床子素得率为评价指标,在单因素试验的基础上,运用Box-Behnken响应曲面法设计料液比、乙醇浓度、提取温度和提取时间4个因素3个水平的试验模型,建立回归方程,优化筛选最佳提取工艺条件。结果表明:模拟得到最佳提取条件为料液比1∶10.28,乙醇浓度77.1%,提取温度72.08℃,提取时间2.5 h。预测蛇床子素得率为13.54 mg/g。为了操作方便,提取条件修正为料液比1∶10,乙醇浓度77%,提取温度72℃,提取时间2.5 h,验证蛇床子素平均得率为13.51 mg/g,与模型预测值较一致。说明响应曲面法优化蛇床子中蛇床子素提取工艺条件,预测值可靠,方法便捷、可行。To study the optimum extract process for osthole from Fructus cnidii, the yield of osthole from the extract was used as the evaluation in- dex, and Box - behnken response surface method was used to design the test models with four factors including material - liquid ratio, ethanol concentration, extract temperature and extract time at three levels based on the single factor experiments, and then the regression equation was established for optimizing and screening the optimum extract conditions. The results showed that the optimum extract conditions were obtained through simulation as follows : material - liquid ratio of 1 : 10.28, ethanol concentration of 77.1% , temperature of 72.08 ~C, and the extract times of 2.5 h. And then the predicted yield of osthole was 13.54 mg/g. In order to facilitate the practice, the conditions were modified as fol- lows : material - liquid ratio of 1 : 10, ethanol concentration of 77%, extraction temperature of 72℃, and extraction time of 2.5 h. The verifica- tion experiment showed the average yield of osthele was 13.51 rag/g, which was more consistent with the predicted value by the modes. The re- suhs indicate that using response surface methodology to optimize the extract conditions for osthole from Fructus cnidii, can obtain a reliable pre- dicted value, it has convenient and feasible advantages.
