摘  要:目的研究根皮苷对仓鼠血清胆固醇水平及肝脏中胆固醇代谢相关调控基因表达的影响。方法试验以饲喂高脂高胆固醇饲料仓鼠为动物模型,随机分为对照组和实验组(0.3%、0.6%和0.9%根皮苷),测定血清总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、甘油三酯(TG)水平。用实时定量荧光PCR(Real-time PCR)和Western blot方法检测肝脏中胆固醇代谢相关调控基因mRNA及蛋白表达水平。结果与对照组相比根皮苷(0.9%)能显著降低血清中TC(P<0.05)和TG(P<0.01)水平;饲喂根皮苷(0.6%,0.9%)能显著升高血清HDL-C水平(P<0.01)。RT–PCR分析显示,肝脏中LDL-R、LXR、CYP7A1基因表达上调,HMG-CoA-R基因表达下调。Western blot结果显示,与对照组相比给予0.6%、0.9%根皮苷后胆固醇合成限速酶HMG-CoA-R蛋白表达水平显著降低(P<0.05),0.9%根皮苷组CYP7A1表达水平显著升高(P<0.05)。结论根皮苷调控机体胆固醇代谢的机制可能是通过调控肝脏中胆固醇代谢相关基因的表达来实现的。Objective To investigate the effects of phlorizin on serum cholesterol level and hepatic cholesterol metabolism genes expression in hamsters fed a high fat and cholesterol diet. Methods In the experiment, 40 hamsters were divided into 4 groups as follows: control group and 3 phlorizin supplemented (PS) groups. Control group was fed a high fat and cholesterol diet, 3 PS groups were fed a high fat and cholesterol diets containing 0.3%, 0.6% and 0.9% phlorizin, respeltively. After 6 weeks feeding, the levels of serum total cholesterol (TC), high density lipoprotein cholesterol (HDL-C) and triacylglycerol (TG) were measured. The mRNA and protein expressions of key enzymes related to cholesterol metabolism were measured by Real-time PCR and Western blot, respectively. Results Serum TC and TG levels were decreased in 0.9% PS group compared with those in control group. In 0.6% and 0.9% PS groups, HDL-C level was significantly increased (P〈0.01) compared with that of control group. RT-PCR assay results showed that the mRNA expressions of LDL-R, LXR and CYP7Alwere increased while the mRNA expression of HMG-CoA-R decreased in the liver. The expression level of protein indicated that HMG-CoA-R in 0.6% and 0.9% PS groups was decreased (P〈0.05) , and CYP7A1 in 0.9% PS group increased (P〈0.05) . compared with those in control group. Conclusion The mechanism underlying the cholesterol-lowering activity of phlorizin is associated with the regulation of hepatic cholesterol-metabolism genes expression in hamsters.
