摘 要:目的:通过构建多囊卵巢综合征(PCOS)大鼠模型,探讨其下丘脑弓状核Kisspeptin/kiss1r系统和促性腺激素释放激素(GnRH)的表达变化及相互作用。方法:孕10dWistar大鼠雌性后代随机分为对照组和PCOS组。采用来曲唑构建大鼠PCOS模型。造模结束后,测定体重参数和内分泌激素的改变;观察卵巢组织的病理学改变;同时用免疫组织化学、免疫印迹法和实时荧光定量PCR检测下丘脑弓状核Kisspeptin、Kiss1r和GnRH的蛋白和各自mRNA水平。结果:成功构建PCOS大鼠模型,体重增加(P<0.05),体脂肪含量增加(P<0.05),血清黄体生成激素(LH)和睾酮(T)水平增加显著(P<0.01),具有典型的PCOS内分泌和卵巢的改变;与对照组相比,PCOS组GnRH的蛋白和mRNA表达显著增加(P<0.01),Kisspeptin蛋白和其mRNA表达下调(P<0.05),未见Kiss1r蛋白和其mRNA表达的显著改变(P>0.05)。结论:大鼠下丘脑弓状核Kisspeptin/Kiss1r系统表达下调,GnRH表达异常增加,与PCOS的神经内分泌有关,可能在PCOS疾病的发生发展中有着重要的作用。Objective: To explore the expressions and interaction between Kisspeptin/Kisslr system and gonadotropin --releasing hormone (GnRH) in the hypothalamic arcuate nucleus of rat models with polycystic ovarian syndrome (P- COS). Methods: Pregnant Wistar rats were obtained and their female pups were divided into the control or PCOS groups randomly. Letrozole was used for established models . Body parameters, endocrine hormones and ovarian mor phology were measured after modeling. Protein expressions and gene levels of Kisspeptin, Kisslr and GnRH in the hy pothalamic areuate nucleus were measured with immunohistochemistry, western blot and qRT--PCR. Results: Rat models with the typical endocrine and ovarian changes in PCOS were successfully established. Compared to the con- trois, the protein expression and gene level of GnRH in the hypothalamie arcuate nucleus of PCOS rats increased sig- nificantly (P〈0.01). Expressions of Kisspeptin/Kisslr system in PCOS rats all decreased (P〈0.01) except the gene level of kiss1 r. Conclusion= Down--regulation of Kisspeptin/Kisslr system and significant increase in GnRH in the hy- pothalamic areuate nucleus of PCOS rats, which could be the results of the endocrine disorder, may play an important role in the development of PCOS.
