摘 要:目的构建以AscI酶切的云南省鼠疫菌PFGE图谱库并探究其流行病学意义。方法采用限制性内切酶AscI对云南家、野鼠型及玉龙鼠疫菌进行酶切分析,并对电泳图谱进行聚类分析。结果30株被试菌株分为15种PFGE型别(相似性系数为88.9%~100.0%)、5个簇、4个群,除404号菌与EV76归为一群,其余3个群分别为家鼠型基因群、野鼠型基因群及玉龙基因群。结论云南鼠疫菌PFGE基因型与生态型相吻合,具有一定的区域聚集性;玉龙鼠疫菌是云南独立的一个基因群。Objective To create fingerprint library of Yunnan Yersinia pest& by pulse field gel electrophoresis (PFGE) with Asc I enzyme digestion and to study its epidemiological significance. Methods We used rare cutting restriction en- zyme Asc I to digest Yunnan E pestis strains isolated from Rattusflavipectus plague focus, .4podemus chevrieri-Eothenomys miletus plague focus and Yulong plague focus, and submitted fingerprints to Bionumerics software for cluster analysis. Re- sults Thirty tested strains were divided into 15 genotypes with 88.9%-100.0% similarity value. 15 genotypes formed 4 groups of 5 clusters, one cluster consisted of No.404 and EV76, the other three were of Rattus flavipectus plague group, Apodemus chevrieri-Eothenomys miletus plague group and Yulong group, respectively. Conclusions PFGE genotypes of Yunnan Y pestis accord with their ecotypes, and have cluster regional feature. The strains isolated from Yulong, are of a unique PFGE type, indicating a new clone group.
