摘 要:利用具有Ri质粒的发根农杆菌 (Agrobacteriumrhizogenes)Pri15834对康乃馨组培苗的带叶茎节进行转化试验。转化后的外植体在K0 附加 50 0mg/L羧苄青霉素培养基上培养 2~ 3周后 ,受感染的伤口部位长出无向地性的毛状根 ,频率为 37.0 7%。将毛状根切碎分别在K0 、K1、K2 培养基上继续培养 3周 ,毛状根在K0 上表现为无限生长 ,在K1上主要表现为长愈伤组织 ,频率为 51.4 2 % ,而在K2 上则主要表现为长绿苗 ,频率为 2 5%。Using Ri plasmid of Agrobacterium rhizogenes (Pri15834), the carnation transformation experiment with leafed stem segments of shoot from tissue culture as initial material was carried out. After transformation, explants were transferred to K 0 medium with 500mg/L carbenicillin and cultured for 2~3 weeks, then non geotropic hairy roots formed from infected wound sites at a frequency of 37.07%. Afterwards,the hairy roots were cut up and subcultured on K 0,K 1,K 2 mediums with 250mg/L carbenicillin for 3 weeks. The results of subculture showed that the hairy roots on K 0 medium grew indefinitely, those on K 1 medium formed calli at a frequency of 51.42%, those on K 2 medium formed green plantlets at a frequency of 25%, indicating expression of Ri plasmid integrated into the genome of carnation cell.
