摘 要:[目的]探讨柠檬醛亚微乳对哮喘大鼠的免疫调节作用及其作用机制。[方法]将60只Wistar大鼠随机分为空白组、模型组、阳性对照组、柠檬醛亚微乳低、中、高剂量组。以卵白蛋白(OVA)致敏大鼠,2周后给予1%OVA雾化激发,在雾化激发前2h用柠檬醛亚微乳剂雾化给药,连续4周。末次雾化攻击24h后,计数肺泡灌洗液(BALF)中白细胞总数和嗜酸性粒细胞(Eosinophil,EOS)数量,测定BALF中IL-4和IFN-γ的含量,并检测肺组织病理改变情况。[结果]与空白组比较,模型组EOS和IL-4显著升高(P<0.01),而IFN-γ显著降低(P<0.01);与模型组比较,阳性组能显著降低EOS数量和IL-4含量(P<0.01),升高IFN-γ的含量(P<0.01),柠檬醛高剂量组EOS数量和IL-4含量显著降低(P<0.01),而IFN-γ的含量则明显升高(P<0.05)。[结论]EOS及细胞因子IL-4和IFN-γ参与哮喘发病过程,柠檬醛亚微乳可通过降低BALF中EOS的数量和IL-4的含量,升高IFN-γ的含量,从而减轻哮喘大鼠的气道炎症。[Objective] To investigate effects and mechanisms of citral submicron emulsion on asthma in Wistar rats. [Methods] Sixty Wistar rats were divided into six groups randomly, which were blank group, model group, positive control group, low, middle, high dose citral submicron emulsion groups. Rats were sensitized with antigen(ovalbumin, OVA). After two weeks, the sensitized rats were chal enged with OVA(10 g&#183;L-1), and before the rats chal enged 2 hours were pretreated with citral submicron emulsion, once per day, every time 30 min, for four weeks. After 24 hours last atomization attack, white blood celldifferential amount, eosinophil amount, the content of IL-4 and IFN-gamma in bronchoalveolar lavage fluid(BALF) were detected. Histological changes of lung tissue were determined. [Results] Compared with blank group, EOS and IL-4 were significantly increased( P〈0.01), while IFN-gamma was significantly decreased in model group( P〈0.01). Compared with model group, the levels of EOS and IL-4 were significantly reduced( P〈0.01), the level of IFN-gamma was increased in positive group( P〈0.01). EOS and IL-4 were decreased significantly in high dose group( P〈0.01), while the content of IFN-gamma was obviously increased(P〈0.05). [Conclusion] EOS, IL-4 and IFN gamma play an important role in the development of asthma. Citral submicron emulsion relieved the airway inflammation in rats by reducing EOS, IL-4 level, and increasing IFN-γlevel possibly.
