摘  要:目的本研究建立了表达人白细胞介素15的重组慢病毒,验证其在人树突状细胞中的表达,希望以此用于疫苗佐剂或基因免疫治疗。方法从人PBMC中扩增IL15基因,克隆至慢病毒表达质粒中,用plvx.ILl5-Ires—gfp包装出重组慢病毒,感染293T细胞用以验证目的基因IL15表达。结果抗生素筛选获得表达IL15的细胞系,目的基因能够在体外稳定的表达。结论构建的慢病毒能够在人未成熟树突状细胞中稳定的表达IL15,可以进一步作为疫苗佐剂或基因免疫治疗使用。Objective To evaluate target gene ILl5 expression by lentiviral vector in mammalian cells, we constructed a recombinant lentivirus expressing human interleukin 15 to be used as vaccine adjuvant or in gene immunotherapy. Methods The ILl5 gene was cloned from human peripheral blood mononuclear cell (PBMC) and constructed into plasmid plvx-IL15-Ires-gfp. Recombinant lentivirus was produced in 293T cells and immature dendritic cells were infected in vitro. Results The target gene expression was detected, cell lines stably expressing ILl5 were obtained by antibiotic screening. Immature dendritic cells can also be infected and express ILl5. Conclusion We successfully constructed lentivirus vector that was able to express ILl5 in dendritic cells without inducing maturation, it could be used in gene immunotherapy or as a vaccine adjuvant.
