摘 要:目的研究模拟失重大鼠灌胃龙血竭后有效成分龙血素B的血浆药物动力学。方法采用3周尾吊大鼠模型模拟失重状态,单次灌胃给予10.6 g/kg龙血竭后,收集各个时间点血浆样本,HPLC-MS/MS法测定尾吊大鼠血浆中龙血素B的含量,并求算药物动力学参数。结果龙血素B在模拟失重大鼠体内血药浓度-时间曲线下面积(AUC0-T)为(23.4±12.4)μg/L·h,达峰时间(Tmax)为(0.33±0.11)h,达峰浓度Cmax(2.77±0.71)μg/L,表观分布容积(Vc)为(677.6±111.6)L/kg。生物半衰期(T0.5)为(8.47±1.7)h。结论本文首次报道了龙血素B在模拟失重大鼠体内的药物动力学参数,为龙血竭对抗空间疾病的相关研究提供了药动学数据。Objective To investigate pharmacokinetics of loureirin B in the plasma of rats under simulated weightlessness after oral administration of Dragon' s Blood. Methods Rats were tail suspended for three weeks to simulate weightlessness. After the rats were orally administered with single dose of 10.6 g/kg Dragon' s Blood, plasma samples were collected and loureirin B was quantified by HPLC-MS/MS method. Pharmacoki- netic parameters were estimated. Results The area under curves (AUC 0-T), time to peak concentration (Tmax ), maximum concentration (Cmax ), volume of distribution (Vc) and biological half-life (T0.5 ) were (23.4 ± 12.4)μg/L · h,(0.33 ±0.11)h,(2.77 ±0.71)μg/L, (677.6 ± lll.6)L/kg, (8.47±1.7)h, respectively. Conclusion The present study firstly reports pharmacokinetic parameters of loureirin B in simula- ted weightless rats, which may provide a pharmacokinetic basis for protective effect of Dragon' s blood on space disease.
