摘 要:为了对芍药再生体系建立进行探究,以芍药无菌苗子叶、茎、叶片为外植体,研究了影响芍药愈伤组织诱导的主要因素。结果表明:诱导愈伤组织较优的外植体为子叶;诱导愈伤组织和增殖的较优培养基为MS+6-BA0.5 mg·L-1+NAA1.0 mg·L-1+2,4-D1.0mg·L-1;芍药器官分化的直接途径优于愈伤组织分化的间接途径,TDZ1.0mg·L-1与NAA0.1 mg·L-1同时使用可提高不定芽的分化率,较优的分化外植体是基部茎。In order to study the way of establishing of regeneration system. stems,cotyledons,leaves of aseptic seedling of peony were selected to study major factors affecting the induction of peony callus. Results showed that the best explants were cotyledons in callus induction. The best medium for callus induction and proliferation was MS+6-BA0.5 mg·L-1+NAA1.0 mg·L-1+2,4-D1.0 mg·L-1;Peony organ differentiation of the direct approach was better than the indirect way of callus differentiation. TDZ with 1.0 mg·L-1 and NAA with 0.1 mg·L-1 used at the same time could increase the rate of adventitious buds differentiation. The best explants of differentiation were the base of stems.
