摘  要:目的:探讨乌梅丸及其寒热配伍对于2型糖尿病(T2DM)大鼠外周组织AMP蛋白激酶[adenosine 5'-monophosphate(AMP)-activated protein kinase,AMPK]表达的影响。方法:采用喂养高脂饮食8周并尾静脉注射小剂量链脲佐菌素(STZ)的方法建立T2DM大鼠模型,乌梅丸及其寒热配伍组分别治疗8周。检测治疗前后大鼠体质量的变化、口服葡萄糖耐量试验(OGTT)、空腹血清胰岛素(FINS)、计算胰岛素抵抗指数(HOMA-IR),分别用Western Blot和RT-PCR法检测肝脏、骨骼肌、脂肪组织中AMPK蛋白和mRNA表达水平。结果:与正常组比较,模型组体质量增加、OGTT明显异常,FINS、HOMA-IR升高,肝脏、骨骼肌、脂肪组织中AMPK蛋白和mRNA表达水平均降低;与模型组比较,乌梅丸及其寒热配伍各组大鼠体质量和OGTT改善,FINS、HOMA-IR下降,肝脏、骨骼肌、脂肪组织中AMPK蛋白和mRNA表达水平明显升高。但乌梅丸组与其寒热配伍各组之间的差异不明显。结论:乌梅丸及其寒热配伍可有效提高肝脏、骨骼肌、脂肪组织AMPK的表达。OBJECTIVE To investigate the effect of Chinese Fructus Mume formula and its separated prescription dismantle- ment study based on warm and cold extracts on the expression of AMP-activated protein kinase in peripheral tissues of type 2 diabetic rats. METHODS The rat model of type 2 diabetes was established by being fed a high-fat diet for 8 weeks and intrave- nous injection of small doses of streptozotocin. The rats in model was divided into five groups: the Fructus Mume formula treat- ment group (FM), the cold property herbs of Fructus Mume formula treatment group (CFM), the warm property herbs of Fructus Mume formula treatment group (WFM), were administrated with Fructus Mume formula and its separated prescrip- tion extracts from a gastric tube, while the rats in diabetic model group (DM) and metformin group (MET) were given by ga- vage with normal saline and metformin correspondingly. The body weight be{ore and after treatment, oral glucose tolerance test (OGTT) and fasting insulin level (FINS) were determined. Homeostasis model assessment-insulin resistance(HOMA-IR)was calculated. The protein and mRNA expression levels of AMPK in liver, muscle and fat tissues were determined by Western blot and RTPCR respectively. RESULTS Compared with the normal group, the animal body weight adding, OGTT, FINS and HOMR IR were all elevated in the diabetic rats, while the protein and mRNA expression levels of AMPK in the liver, muscle and fat tissues were markedly decreased in model group. Compared with the model group, the body weight adding, OG- TT, FINS and HOMR-IR were all reduced in the rats of Fructus Mume formula and its separated prescription extracts groups. Meanwhile, the protein and mRNA expression levels of AMPK in the liver, muscle and fat tissues were markedly increased in Fructus Mume formula and its separated prescription extracts groups. CONCLUSION Fructus Mume formula and its separa- ted prescription extracts could be effective on treating type 2 diabetes in rats. The mechani
