摘 要:β-氨基己糖苷酶(β-Hexosaminidase,β-Hex,EC 3.2.1.52)是生物界广泛存在的一种糖苷水解酶。通过全基因组信息鉴定甜瓜β-氨基己糖苷酶基因家族,初步在甜瓜全基因组数据库中发现5个β-Hex基因成员,采用Pfam等在线软件预测候选蛋白的结构域,去除2个不完整的基因,最后将筛选出的3个基因与拟南芥、青椒、番茄、可可树、谷子、葡萄和黄瓜的相应基因编码的蛋白序列进行进化分析比对、建立系统发生树和保守基序的预测分析。结果表明,进化树亚族成员中基序相同,并且在保守区序列高度相似;亚族之间基序差异也较小,保守区序列也具有较高相似性。β-Hexosaminidase ( β-Hex, EC 3.2.1.52) is one of glycosidases, have been shown to be universally distributed among most types of living organisms. β-Hexosaminidase family genes in Cucumis melo were genome-wide analyzed and 5 members of β-Hex family genes were found in the melon genome database. With the analysis of protein structure prediction software such as Pfam, we removed 2 incomplete genes and filtered out 3 genes in C. melo. These protein sequences were analyzed by multi-sequence aligned, which were similar in Arabidopsis thaliana, Capsicum annuum, Solanum lycopersicum, Theobroma cacao, Setaria italica, Vitis vinifera, C.sativus and C.melo. The structure evolution relation and conserved motifs were researched by bioinformatics method. The results indicated that this gene family was highly conservative with same motifs in the same β-Hex family. Each β-Hex family within phylogenesis tree demonstrated a bit of differences and the maior domain were conserved too.
