抗生素对巴西橡胶树胚性悬浮细胞生长的影响

摘　　要：为了评估不同浓度抗生素对胚性悬浮细胞生长的影响,将橡胶树胚性悬浮细胞接种到不同浓度的替卡西林和卡那霉素的培养基上,结果表明,添加0～500 mg/L的替卡西林对胚性悬浮细胞生长影响较小;添加50 mg/L的卡那霉素能抑制胚性悬浮细胞生长,添加100 mg/L的卡那霉素导致部分胚性悬浮细胞死亡.将经共培养的胚性悬浮细胞接种到添加不同浓度普卡西林的培养基上,结果发现,300mg/L的替卡西林能较好的抑制农杆菌的增殖.替卡西林和卡那霉素可分别作为农杆菌介导的转化橡胶树胚性悬浮细胞的抑菌和筛选抗生素,替卡西林浓度以300 mg/L较好;卡那霉素50～10o mg/L较好.In order to evaluate effects of different concentrations antibiotics on embryogenic suspension cell growth,the embryogenic suspension cells were inoculated into media supplemented with different concentrations of ticacillin and kanamycin.The results showed that the growth of embryogenic suspension cells was less affected with 0-500 mg/L ticarcillin in medium,the growth of embryogenic suspension cells was inhibited with 50 mg/L kanamycin in medium,and some embryogenic suspension cella led to death when supplemented with 100 mg/L kanamycin in medium.Embryogenic suspension cells after co-culture were inoculated into media added with different concentrations of ticarcillin,which showed that proliferation of Agrobactrium could be better suppressed with 300 mg/L ticarcillin in medium.In conclusion,ticarcillin was a good antibiotic to eliminate Agrobactedum tumefaciens in Agrobaaerium-mediated transformtion of embryogenic cell suspension of Hevea brasiliensis with an optimal concentration of 300 mg/L,and kanain ycin 50-100 mg/L.
