摘  要:目的:建立慢性心力衰竭大鼠模型,并对模型大鼠气虚证和血瘀证相关信息进行客观评价。方法:采用左冠状动脉前降支高位结扎法复制大鼠心力衰竭模型,术后多个时间点,动态观察大鼠宏观体征,计算大鼠应激后呼吸频率,采集大鼠足底图像进行色度分析,同时术后在第27-30天对大鼠分别进行游泳力竭实验、旷场实验、超声心动检测、血流动力学检测。结果:模型组大鼠从术后第7天开始,各时间点模型组大鼠应激后呼吸频率较假手术大鼠均明显增加(P<0.05,P<0.01),足底R、G、B值较假手术组大鼠均降低(P<0.05,P<0.01);术后第27天模型组旷场实验总运动距离和平均运动速度明显下降(P<0.01);术后第28天模型组游泳力竭时间缩短(P<0.01);术后第29天超声心动数据和术后第30天血流动力学检测数据表明模型组大鼠心功能下降。结论:左冠状动脉前降支高位结扎法可以复制慢性心力衰竭模型,并且该模型在术后第7-28天可辨证为气虚血瘀证。Objective: To establish the model of chronic heart failure in rats, and to evaluate qi deficiency syndrome and blood stasis syndrome objectively. Methods: SD rats were subjected to either proximal left anterior descending coronary artery (LAD) ligation or sham operation. Global physical signs observation, colorimetric analysis on images of plantar, the respiratory rate post-stress were counted in every time point. After 27-30 days postoperatively all the rats were subjected to forced swimming test (FST) to measure the exhaustive swimming time, open field test, ultrasonic echocardiography, hemodynamics and pathological methods. Results: From the 7th day postoperatively, compared with the sham operation group, model rats' respiratory rate was faster (P〈0.05, P〈0.01), and shared a lighter degree of RGB valve, which meaned they had much purple dark skin colors. Exhaustive swimming time were shorter (P〈0.01), total distance moved were shorter (P〈0.01), velocity were slower and the cardiac function of rats was significant decreasing in model group. Conclusion: Chronic heart failure induced by LAD ligation can be diagnosed as the syndrome of qi-deficiency and blood stasis between 7-28 days after the surgery.
