基于小鼠胚泡着床障碍模型探讨二补助育汤对子宫内膜SCF mRNA表达的影响

摘　　要：目的:观察二补助育汤对胚泡着床障碍模型小鼠子宫内膜SCF mRNA表达的影响。方法:建立小鼠胚泡着床障碍模型,将实验动物分为7组:空白组,模型组,补佳乐组,阿司匹林组,中药低、中、高剂量组。于造模的同时给药后处死小鼠,检测小鼠子宫内膜SCF mRNA的表达量。结果:模型组小鼠子宫内膜SCF mRNA的表达明显低于空白组(P<0.05);与模型组比较,中药各治疗组及西药各组小鼠子宫内膜SCF mRNA表达明显增加(P<0.05),中药高、中剂量组子宫内膜SCF mRNA表达明显高于低剂量组(P<0.05)。结论:SCF可能通过作用于其受体C2KIT,从而使胚泡能更好的黏附于子宫内膜,利于其着床;二补助育汤改善子宫内膜容受性的机制可能是通过调控子宫内膜SCF mRNA的表达来实现,从而使卵泡发育与子宫内膜同步,利于胚泡着床。Objective： To investigate the effects of Erbuzhuyu Decoction in SCF mRNA expression based on the mouse model of embryo implantation dysfunction. Methods： To establish mouse model of embryo implantation dysfunction, divide model animals into seven groups： blank group, model group, western medicine groupl （Progynova）, western medicine group2 （Aspirin）, low-dose Erbuzhuyu Decoction group, middle-dose Erbuzhuyu Decoction group, high-dose Erbuzhuyu Decoction group. Those groups were modeled and administrated before detecting the expression of SCF mRNA. Results： The expression of SCF mRNA of the model group was significantly lower than the control group; Compared with the model group, the expression of all the positive groups and the Erbuzhuyu Decoction groups significantly increased （P〈0.05）, showed that the positive samples and the Erbuzhuyu Decoction can promote the expression of SCF mRNA; Middle-dose Erbuzhuyu Decoction group and high- dose Erbuzhuyu Decoction group have more obvious effects than low-dose Erbuzhuyu Decoction group expression of SCF mRNA （P〈0.05）. Conclusion： SCF can act on C2KIT by helping the embryo adhesion, which is conducive to embryo implantation; Erbuzhuyu Decoction may improve endometrial receptivity by regulating SCF mR_NA expression so that embryo can grow with the endometrium, which is conducive to embryo implantation.
