摘　　要：目的:建立采用UPLC-TQ-MS分析法同时测定补肾活血方中儿茶素、芍药苷、金丝桃苷、阿魏酸、虎杖苷、槲皮素、白藜芦醇、补骨脂素、异补骨脂素、淫羊藿次苷Ⅱ、蛇床子素、异欧前胡素含量的方法。方法:采用ACQUITY UPLC BEH C18(2.1mm×100mm ID,1.7μm,Waters,Milford,USA)色谱柱,乙腈-0.1%甲酸溶液为流动相梯度洗脱;样品经电喷雾离子源正负离子化后,通过Waters-Xevo TQ三重四级杆串联质谱仪,采用多反应离子监测(MRM)对12种化合物进行含量测定。结果:复方水提液中儿茶素、芍药苷、金丝桃苷、阿魏酸、虎杖苷、槲皮素、白藜芦醇、补骨脂素、异补骨脂素、淫羊藿次苷Ⅱ、蛇床子素、异欧前胡素含量分别为0.0422、4.2596、0.1211、0.0584、0.7594、0.2001、0.1570、0.8948、0.4634、0.0928、0.5364、0.1487mg/g。结论:采用LC-MS/MS分析法同时测定复方中12种有效成分含量,该方法快速、便利,结果准确,对于研究补肾活血方配伍前后化学成分含量的变化,探究复方药效活性的物质基础具有重要意义。Objective： To establish a method of UPLC-TQ-MS for the content determination of Catechin, Paeoniflorin, Hyperin, Ferulic acid, Polydatin, Quercetin, Resveratrol, Psoralen, Isopsoralen, Icariside II, Osthole and Isoimperatorin in Bushen Huoxue Formula. Methods： The content determination of the 12 compounds was performed on an ACQUITY UPLC BEH C18 （2.1mmxl00mm ID, 1.7p_m, Waters, Milford, USA） chromatographic column （mobile phase was HCN-0.1%H3PO4）. Waters- Xevo TQ Triple-stage Quadrupole mass spectrometry system equipped with an electrospray ionization source was used as the detector and operated in the negative ion mode. Multiple reactions monitoring （MRM） was performed to detect the 12 compounds. Results： The contents of 12 compounds were 0.0422, 4.2596, 0.1211, 0.0584, 0.7594, 0.2001, 0.1570, 0.8948, 0.4634, 0.0928, 0.5364, 0.1487mg/g in the water extract of Bushen Huoxue Formula respectively. Conclusion： The method was accurate, simple and replicable for studying the content of the 12 compounds in Bushen Huoxue Formula compatibility. The result is of great significance for studying the change and the content of chemical components, before and after compatibility of Bushen Huoxue Formula, also has an important reference significance for the substance basis research.
